Objective:In this study,chicken embryos were used as raw materials to prepare enzymatic hydrolysates of chicken embryos and their antioxidative activities were investigated.Chelates of chicken embryo peptides and trace elements zinc were prepared and their anti-fatigue and immunity-improving functions were investigated.Methods:The optimal enzymolysis process of chicken embryo eggs was optimized by single factor test and orthogonal test.Activated charcoal was used to decolorize the chicken embryo peptides prepared by the enzymatic hydrolysis process and use ultrafiltration to segment them.The best chelation process was obtained by the single factor test method and the orthogonal test method for the chelation process of chicken embryo peptide and trace element zinc.Finally,a preliminary anti-fatigue and immunomodulatory pharmacodynamics test was performed to verify the effect of the zinc-rich chicken embryo peptide.Results:The best enzymolysis method for chicken eggs is to use trypsin with alkaline protease first,the best enzymatic process:substrate concentration is 10%,temperature is adjusted to 55?,pH is 8.0,3.0%alkaline proteolysis was carried out for 3h until the temperature dropped to 40?.After adding 2.0%trypsin for 2h,the hydrolysis degree of the enzymatic hydrolysis process was 31.96%.The best deodorization and decolorization process is to add 0.5%powdered activated carbon under the condition of 10%substrate concentration,adjust the pH to 5.0,heat and stir in a water bath at 65?for 40 minutes,the decolorization rate is 88.57%,the peptide recovery rate is 82.63%.It has a small taste.The results of the segmentation and antioxidant activity of chicken embryo peptides showed that the anti-oxidation activity of chicken peptides with a molecular weight of<1KDa was the best,IC50 was 0.25 mg/ml.The best method of zinc chelation in zinc-rich chick embryo peptides is as follows:The chelating temperature is selected at 70?,the ratio of zinc ion to chicken embryo peptide concentration is 1:2,the pH is 8.0,and the chelating time is 2h under this reaction condition,the chelation rate was 52.63%,and the content of zinc in this chelate was determined to be 0.0937%by flame atomic absorption spectrometry.After preliminary pharmacodynamic study,it was found that zinc-rich chicken embryo peptide has better anti-fatigue and immune-enhancing effects.Conclusion:The chicken embryo peptides prepared by the enzymatic hydrolysis of the two enzymes have good antioxidant activity;the zinc-rich chicken embryo peptides prepared after chelation of the chicken embryo peptides with zinc have better anti-fatigue and immunity-improving capabilities.
