Objective:The model of acute rat liver injury induced by-10? low temperature environment was established to simulate the actual military transport and operational rules.At the same time,we want to explore the preventive and therapeutic effects of Cistanche deserticola on acute liver injury induced by low temperature and possible mechanisms.Methods: Establishing the model of acute rat liver injury induced by-10? low temperature environment.Studying the mechanism of acute liver injury induced by hypothermia in rats with detecting ALT,AST,SOD,MDA,TNF-a,IL-10 and Caspase-3 in the serum and observing the pathological changes in rat liver.Exploring the preventive and therapeutic effects of Cistanche deserticola on acute liver injury induced by comparing the difference of liver injury between high dose Cistanche deserticola group? Cistanche deserticola low dose group and Silibin Meglumine Tablets group.Results:(1)Liver function:Compared with the control group,the difference was obvious in model group(P<0.01),suggesting that the model was successful;Compared with the model group,low dose group of Cistanche deserticola in each time period were not statistically significant(P>0.05),and there are some difference about high dose Cistanche deserticola group and Silibin Meglumine Tablets group at 2 hours and 4 hours(P<0.05).(2)Oxidative stress:At the three time period,the change of SOD activity and MDA content in model group is obvious than the control group(P<0.01),suggesting that the model was successful;Compared with the model group,the activity of SOD about high dose Cistanche deserticola group and Silibin Meglumine Tablets group have statistically significant(P<0.05);The content of MDA in high dose Cistanche deserticola group and Silibin Meglumine Tablets group is slightly lower than the model group at 2 hours(P<0.05),there are some difference with high dose Cistanche deserticola group and model group about the content of MDA at 4 hours and 6 hours(P<0.05)(3)Inflammatory factors:Compared with the control group,changes of TNF-a and IL-10 in model group was clear(P<0.01),suggesting that the model was successful;Compared with model group,high dose Cistanche deserticola group and Silibin Meglumine Tablets group about the indexes were different at 2 hours(P<0.01,P< 0.05);In the drug groups the content of TNF-a is decreased(P <0.05),while the content of IL-10 in high dose Cistanche deserticola group and Silibin Meglumine Tablets group is increased than model group at 4 hours(P <0.05);Only the high dose Cistanche deserticola group has an intervention in the indexs(P < 0.05).(4)Hepatocyte apoptosis: The content of caspase-3 in the model group is higher than the control group(P<0.01),suggesting that the model was successful;The content of caspase-3 in high dose Cistanche deserticola group and Silibin Meglumine Tablets group is decreased than model group at 2hours and 4 hours(P<0.05);The content of caspase-3 in high dose Cistanche deserticola group is much little than model group(P<0.05).(5)Pathology: With the prolongation of cold exposure time,liver injury is aggravating,but compared with model group,Cistanche deserticola group and Silibin Meglumine Tablets group can improve the injury of liver.Conclusion: 1.A rat model of acute liver injury induced by low temperature environment is established,which is easy to operate and replicated.2.Cistanche deserticola has protective effect on acute liver injury induced by low temperature environment.The mechanism may be related to the increasing of SOD activity,the reduction of inflammatory reaction and the inhibition of hepatocyte apoptosis.
