The endosperm of Gleditsia sinensis Lam.is rich in nutrients,nourishing heart and promoting pulse,clearing liver and improving eyesight,strengthening spleen and nourishing kidney,eliminating phlegm and inducing resuscitation.It is a valuable natural green nourishing food to harmonise functions of human organs.Currently,development and utilization of some nutrients in the endosperm of Gleditsia sinensis Lam.are still not clear and need further study.The single factor and response surface method were used to optimize the extraction technology of crude polysaccharides from the endosperm of Gleditsia sinensis Lam,and the immune activity of the endosperm of Gleditsia sinensis Lam.polysaccharides was evaluated by mice as experimental animal models,which peovided the basic theoretical basis for the development of the endosperm of Gleditsia sinensis Lam.products.The effects of temperature,time and ratio of liquid to material on the rehydration of the endosperm of Gleditsia sinensis Lam.were investigated by single factor experiment.The rehydration conditions were determined as follows:rehydration temperature 80 ℃,rehydration time 30 min,liquid to material ratio 40 mL/g.Experiments were carried out by thermal extraction of single factor and response surface experiments to determine the optimization extracting tradition of crude polysaccharides from the endosperm of Gledit.sia sinensis Lam.:extraction temperature 93 ℃,extraction time 1.7 h,extraction liquid ratio 180 mL/g,extracted 2 times.Under these optimal conditions,the crude polysaccharide extraction rate was 57.54%,the solvent was evaporated by rotary vaccum and concentrated to 1/2 of its original volume,then precipitated by the addition of 2.8 volumes of 95%ethanol and stirred rapidly,precipitated 4 ℃ for 12 h.The final precipitation was then dried under 60℃ which obtain crude polysaccharides 86.88 mg/g.The mice were used as experimental animal models to study the immune activity of the endosperm of Gleditsia sinensis Lam.crude polysaccharides.In the present study,we have attempted to illustrate efficacy of crude polysaccharides in CY-induced immunosuppressive mice which were treated with the crude polysaccharides.The result of experiment showed that crude polysaccharides under the doses of 50 mg/(Kg·bw)、100 mg/(Kg·bw)and 150 mg/(Kg.bw)can improve the body weight,rgan index of thymus and spleen.Meanwhile,crude polysaccharides could also increase the activity of phagocytosis of peritoneal macrophages,NK cytotoxicity and the proliferation level of splenic lymphocyte.In addition,the different dose of crude polysaccharides could significantly enhance the activity of GSH-PX,SOD,AKP and CAT of blood serum and liver.Through the method of PCR,we found that the crude polysaccharides could enhance the experession of IL-2、IL-6、IFN-γ.Mcl-1 and Bcl-xL which related with immunity.
