Objective : bladder cancer is the most common malignant tumor of the urinary system,accounting for the first place in the incidence of urogenital system tumors in China,and is about to become the sixth most common malignant tumor in the world,and the incidence is still rising.It has the characteristics of rapid metastasis,poor prognosis and high recurrence rate.Bladder cancer is a malignant tumor that occurs on the mucosa of the bladder.According to histopathological classification,the pathological types of bladder cancer include bladder urothelial carcinoma,bladder adenocarcinoma,bladder squamous cell carcinoma,bladder small cell carcinoma,bladder carcinoid carcinoma and bladder transparent cell carcinoma.Among them,bladder urothelial carcinoma accounts for more than 90% of the total number of bladder cancer patients,and is the most common bladder cancer.Although localized bladder cancer can be controlled by surgical resection,there is still a lack of effective treatment for the high recurrence and metastasis rate of bladder cancer after surgery.Therefore,how to prevent the development and progression of bladder cancer has become a key problem to be solved in the treatment of bladder cancer,among which,the search for new molecular targets and the exploration of the molecular mechanism of bladder cancer development have become a research hotspot.Gene mutation,loss of tumor suppressor and abnormal regulation of cell growth signaling pathway are all related to bladder cancer.In recent years,a large number of studies have shown that the abnormal expression of non-coding RNA(ncRNA)plays an important role in the occurrence and development of bladder cancer.Therefore,bioinformatics database was used in this study to study the genes(mRNA,miRNA and lncRNA)in bladder cancer from the molecular level,so as to enrich the occurrence and development mechanism of bladder cancer and provide effective measures and methods for the clinical prevention and prognosis improvement of bladder cancer.Methods: bioinformatics is an interdisciplinary subject developed in recent years.It covers all aspects of the acquisition,processing,storage,distribution,analysis and interpretation of biological information,and integrates various technologies such as medical science,biostatistics,statistics,informatics and computer science.Biological analysis of large data such as high-throughput sequencing technology and popularization,it is first filtered for biochip large amounts of data,and USES the statistical analysis,sequence alignment,path analysis,and visualization mapping,to integrate data mining and analysis of diseases on molecular level,extension of disease progression(especially in molecular level)in-depth understanding.With the development of bioinformatics,the research model of biology is gradually mature,that is,to collect and use the existing data information,make theoretical speculation first,and then through data analysis and experimental verification.This study based on the research of the TCGA bioinformatics database,using the Illumina HiSeqRNASeq and Illumina HiSeqmi platform to generate transcription genome(RNA-seq)data,using R software screen differentially expressed in bladder cancer RNA,combined a series of bioinformatics software and mining methods such as literature,analyzing the interaction relationship between them so as to explore the differentially expressed genes related to bladder cancer pathogenesis,to better understand the molecular mechanism of occurrence,development and prognosis of bladder cancer provides important molecular information,It provides a new direction for further study on the early diagnosis,treatment and prognosis of bladder cancer.Results: a total of 1819 mrnas,659 lncrnas and 160 mirnas were identified to be significantly differentially expressed in BC,among which 52 mrnas,58 lncrnas and 22 mirnas were incorporated into the ceRNA network we initially constructed.Both CFL2 and TPM2 were down-regulated in BC and showed significant correlation.HOXB5 and 6 lncrnas(adamts9-as1,AC112721.1,LINC00460,AC110491.1,LINC00163,HCG22)were closely related to high grade,disease stage and overall survival.Conclusion: in this study,we have screened out differentially expressed mRNA,lncRNA and miRNA that are closely related to tumorigenesis and prognosis in BC.Further experimental studies are needed to verify these results.
