Objective:To investigate the effection of enhancing the memory and eliminating the free radical of the mixture which contains seven different kinds of nuts and dried fruits from Xinjiang area to the rats that had been modelize to AD.Methods:Deviding the 75SPF mice into 5 groups randomly?15 rats in each group?:shame operation group?Control group?low/medium/high dose group.We injected A?25-355-35 to the both sides of CA1 area of hippocampus of model groups mice to make the AD mice models,?The shame operation group injected the same volume of SPSS?.And feed the each groups of mice with food that contains different concentration of nuts mixture?or food without nuts and dried fruits mixture?for 6 weeks.And we did Y-puzzle?started in 5th weeks?and Morris water puzzle?started in 6th weeks?experiments to test the studyingand memorizing ability of mice in different groups.then we collect the sample of blood serum and hippocampus tissue,HE staining sections were made to observe the pathological change of CAI area.Using ELISA to measure the concentration of malonaldehyde that sample contains and the activity of catalase and SOD,By which we could analyze the Effects of dried fruit diet on oxidative stress in AD model rats.Results:?1?behavioral experiment:compared with the sham operation group,the escape latency of y-type maze in rats in the model control group was significantly increased?P<0.05?,the number of error response?EN?was significantly increased?P<0.05?,the number of all-day response?TRT?was significantly increased?P<0.05?,and the number of days required to reach the standard was significantly increased?P<0.05?.Morris is the directional navigation experiment of maze experiment,the incubation period of the model control group increased significantly?P<0.05?,and the number of target quadrant entry decreased significantly?P<0.05?.In the space exploration experiment,the incubation period of the model group decreased?P<0.05?,the original platform quadrant retention time and the number of times of crossing the original platform area all decreased significantly?P<0.05?,and the difference was statistically significant.?2?Observation results of HE stained sections:Compared with the sham operation group,the neurons in the CA1 area of the hippocampus in the other groups were damaged and pathological changes occurred,and the pathological changes were the most serious in the model group.Compared with the model group,the neurons in the high concentration group were significantly improved.Results of oxidative stress experiment:Compared with the sham operation group,MDA content in serum and hippocampal homogenates of the other groups was significantly reduced?P<0.05?,and SOD and CAT activity were significantly increased?P<0.05?.Compared with the model group,MDA content was significantly decreased in the high concentration group?P<0.05?,CAT activity was significantly increased in the medium and high concentration groups,and SOD activity was significantly increased in the low,medium and high concentration groups?P<0.05?.In the hippocampal homogenate,MDA content was significantly decreased in the medium and high concentration groups?P<0.05?,CAT activity was significantly increased in the high concentration group,and SOD activity was significantly increased in the low and high concentration group?P<0.05?.Conclusion:?1?The combination of dried fruits and nuts can improve the memory and learning ability of AD rats,and can protect AD to a certain extent.?2?The protective effect of a combination of dried fruits on AD may be related to Enhancing oxidative stress ability in AD rats,At the same time,it may be related to the combination may improve the damage of neurons in hippocampal CA1 region of AD rats.
