Jin Lian Hua is the dried flower of Trollius chinensis Bunge belong to the ranunculaceae family,which has antibacterial,antiviral,antitumor and antioxidant biological activities.It is often used for the treatment of respiratory tract infection,pharyngitis,tonsillitis and bronchitis.In recent years,the studies on the chemical constituents and pharmacological effects of T.chinensis were mainly focused on the two flavone C-glycoside orientin and vitexin,but the other constituents in T.chinensis were seldom studied.Besides,there is still no unified evaluation standard for the quality of Trollius chinensis Bunge.Therefore,the dried flowers of T.chinensis was chosen as the research object of our study.21 compounds were isolated from the 70%ethanol extract of T.chinensis by various chromatographic techniques,such as silica gel column chromatography,sephadex LH-20 column chromatography,gel column chromatography,reverse column chromatography,medium and low pressure column chromatography,recrystallization,preparation and semi-preparative HPLC,etc.Their structures were elucidated by HR-MS,NMR spectroscopy.Among them,4 and 13 are phenolic acids,while compound 4 is a new compound.The remaining 19 compounds are all flavonoids,among which,5,14,15,18 and 19 are new compounds.1,16 are flavonoid O-glycosides,which were isolated for the first time in this genus.Compounds 2-3,5-12,14-15,17-21 are flavonoid C-glycosides,and compounds 14,15,18,19,21 are relatively rare flavonoid lactosides.In addition,based on the experiments of staphylococ cus aureus growth inhibition model,DPPH radical scavenging and NF-?B reportor gene assays,the antibacterial,antioxidant and anti-inflammatory activities of 21 compounds were evaluated,and the preliminary structure-activity relationship were discussed:flavonoid C-glycosides showed stronger biological activity than flavonoid O-glycosides as well as flavonoid lactosides have better biological activities compared with other glycosides in antibacterial and antioxidant aspects.The above studies have preliminarily clarified the material basis of the treatment of respiratory infections in the clinical use of T.chinensis,and provide a basis for further rational development of T.chinensis.In this paper,the fingerprint of the extract of T.chinensis was further established,and the chromatographic peaks of the isolated compounds were identified.Besides,the mass spectrometry fragmentation of the isolated monomeric compounds was summarized to detect the other flavonoid C-glycosides from the T.chinensis extract.A total of 37 flavonoid glycosides were identified,and the structural types of 20 flavonoid glycosides were preliminarily speculated,including 8 flavonoid lactosides and 12 flavonoid glucosides.This study laid a foundation for a more in-depth study of the chemical constituents in this plant and provided a reference for the establishment of quality control methods of T.chinensis.
