Jilin Province is one of the main distributive areas of Rhodiola sachalinensis,as a herbal medicinal plant,Rhodiola sachalinensis has been shown to effect on the human body in anti-anoxia,anti-fatigue and anti-tumor.In recent years,because the incidence of prostate cancer has increased significantly,the early symptoms of prostate cancer are not obvious,and factors such as advanced stage have been diagnosed at the time of diagnosis,the research on prostate cancer has become the focus.The early treatment of prostate cancer mainly adopts surgical treatment,and castration in the later stage,however,due to its drug resistance in this stage,changing from androgen dependence into androgen non-dependence,finally,only treated with chemotherapy.For the past few years,due to the non-toxic and non-irritating advantages of the natural plant to human body,it has been more and more valued in the study of tumor.The data shows that more than 60%of FDA approved anti-cancer drugs roots in natural compounds,including medicinal plant.So the PC3 and LNCaP cells of human prostate cancer were used as research object in this experiment to study the mechanism of action that irradiated Rhodiola sachalinensis extract to the proliferation and apoptosis of PC3 and LNCaP cells of prostate cancer.In this experiment,Rhodiola sachalinensis was sent to Sichuan Institute of Atomic Energy for irradiation with the dose 5,10,20,and 30 kGy,respectively,and 60Co-? ray were used for irradiation.Then use the Rhodiola sachalinensis extract in vitro anti-cancer test.The CCK-8 method was adopted to detect the effect of irradiated Rhodiola sachalinensis extract on PC3 and LNCaP cells activity in the experiment,and the morphological changes of PC3 and LNCaP cells after the action of 20 kGy irradiated Rhodiola sachalinensis extract were observed by inverted microscope,also the migration ability of the two kinds of cells by scratch-wound experiment.The effect of drugs on the two kinds of prostate cancer cell periodic distribution and apoptosis was further detected by FACS,and the expression of related apoptin PCNA and Bcl-2 within PC3 and LNCaP cells after the treatment of 20 kGy irradiated Rhodiola sachalinensis extract was detected by Western blot technology.The experimental results showed that both the irradiated Rhodiola sachalinensis extract and that of non-irradiated could inhibit the proliferation of the two kinds of prostate cancer cells after 48h-action on PC3 and LNCaP cells,the inhibitory effect of irradiation Rhodiola sachalinensis extract stronger than that non-irradiated group,among which had the highest inhibitory rate of irradiated 20 kGy Rhodiola sachalinensis extract.The high,medium and low concentrations of 20 kGy irradiation dose group were detected respectively 300 ?g/mL,600 ?g/mL,900 ?g/mL to PC3 cells by CCK-8 experiment,and those of LNCaP cells were respectively 200?g/mL,400?g/mL,800?g/mL.In the experiment,compared with the control group,the high,medium and low concentrations of 20 kGy Rhodiola sachalinensis extract inhibited the migration of PC3 and LNCaP cells in prostate cancer,and 900 ?g/mL and 800 ?g/mL group had the best inhibitory migration effect in PC3 and LNCaP cells.The periodic and apoptosis result of FACS detection of prostate cancer PC3 and LNCaP cells showed that the cell cycle could be blocked after action-on PC3 and LNCaP cell 48 h by both the 20 kGy irradiated Rhodiola sachalinensis extract,meanwhile,followed by the increase of drug action concentration,the apoptosis rate of two kinds of prostate cancer cells tended to increase.Western blot result showed that both the 20 kGy irradiated Rhodiola sachalinensis extract could reduce the anti-apoptosis-related protein PCNA and Bcl-2 with dose-dependence.In conclusion,the irradiated Rhodiola sachalinensis extract group could significantly inhibit the proliferation of prostate cells compared with the non-irradiated group,and it was possible to inhibit the proliferation action of prostate cancer by inducing the cell apoptosis and reducing related inhibitor of apoptoasis protein through the migration of prostate cancer PC3 and LNCaP cells.
