Objective(1)To detect the serum level of cytokines interleukin(IL)-33 and sST2 in patients with non,segmental generalized vitiligo onset period of times and investigate the correlation between IL-33、sST2 and non-segmental generalized vitiligo.(2)To detect the expression in peripheral blood mononuclear cells of IL-33mRNA and sST2mRNA transcription level in patients with non-segmental generalized vitiligo onset period of times and observe the possible role of it in the non-segmental generalized vitiligo.Method(1)Peripheral blood was collected from patients with non-segmental generalized vitiligo in a total of 54 cases,including progressive(28 cases),stationary(26 cases)and 30 healthy human controls.All cases will be divided into three groups:the progressive stage group,stationary group and control group.To examine the levels of IL-33 and sST2 in the serum,we used enzyme-linked immuno sorbent assay(ELISA)to do the experiment.(2)The expression of IL-33mRNA and sST2mRNA in peripheral blood mononuclear cells was detected by reverse transcriotion-polymerase chain reaction(RT-PCR).Results(1)The level of serum IL-33 was higher in the progressive stage and stationary stage of non-segmental generalized vitiligo than in the normal controls,the progressive group compared with the control group,the difference was statistically significant(t=7.079,p<0.05),while the stationary group compared with the control group respectively,the difference was no statistically significant(t= 1.081,p>0.05).(2)The patients with progressive group and stationary group of non-segmental generalized vitiligo showed a higher serum level of sST2 compared with the healthy controls,the progressive group and the stationary group compared with the control group,the difference was statistically significant(t=9.329、t-2.999,p<0.05).(3)The IL-33mRNA expression quantity of PBMCs in the progressive stage and stationary stage of non-segmental generalized vitiligo was separately 4.78、4.23times of healthy controls.(4)The sST2mRNA expression quantity of PBMCs in the progressive stage and stationary stage of non-segmental generalized vitiligo was separately 2.1.71 times of healthy controls.Conclusion The levels of serum IL-33 and sST2 were significant higher in the non-segmental generalized vitiligo than in the normal controls,the experimental data show that the progressive group is more significant than stationary group,the stationary group compared with the control group,the difference was not statistically significant,it means that it play a major role in the pathogenesis and disease activity of vitiligo.The results of RT-PCT shows that IL-33mRNA and sST2mRNA expression of PBMCs in non-segmental generalized vitiligo is increased compared with the control group and the difference was statistically significant,it suggests that IL-33 and sST2 may be closely associated with the pathogenesis of non-segmental generalized vitiligo.
