Purpose:Rosmarinci acid(RA),a natural water-soluble polyphenol,exsist in lots of aromatic plants,have a variety of potential therapeutic properties.Including antioxidant,anti-apoptosis,anti-fibrosis,liver protection,anti-neurodegeneration and anti-inflammatory activity.The study destination is to explore the preventive effect of RA in Diethylnitrosamine(DEN)-induced primary hepatocellular carcinoma(PHC)in rats.Method:Constructing a rat model of primary hepatocellular carcinoma induced by DEN.Rats were randomly divided into control group,DEN group,CTX group,and RA(100,200 mg/kg)group.They were delivered drugs once daily for 6 weeks,then collecting blood and liver sample to testing blood routine and blood biochemistry targets of rats;HE was used to observe Pathological changes;Method elisa was be taken to detect the level of AFP in liver;Method immunohistochemical was be taken to detect the expression of NF-?B p65?TGF-?;Method real-time fluorescent quantitative PCR was be taken to detect the mRNA expression of TNF-? and IL-10 in liver.Result:Compared with the control group,liver samples from DEN rats had obviously increased the level of ALT,AST and TBIL(P<0.05).Compared with DEN group,giving RA treat with 100,200 mg/kg dose can low down the leve of ALT,AST and TBIL(P<0.05).The antioxidant experiments of rats indicated that RA 100,200 mg/kg treatment increased rhe target SOD and reduce the target MDA compared with DEN-induced model group(P<0.05).Target HE showed giving RA treat with 100,200 mg/kg dose clearly improved histopathological state of DEN-induced primary hepatocellular carcinoma.Compared with the model group,giving RA treat with 100,200 mg/kg dose can reduce the expression level of AFP(P<0.05);Giving RA treat with 100,200 mg/kg dose can inhibit the process of phosphorylation of NF-?B and reduce the expression level of TGF-? immunohistochemistry.Compared with DEN group,RA dose of 100 mg/kg or 200 mg/kg clearly decrease the mRNA level of TNF-? and IL-10 in the real-time fluorescence quantitative PCR experiment.Conclusion:RA had Preventive effect in DEN-indeced primary hepatocellular carcinoma on rats.The effects may by interdicting NF-?B pathway to decrease the expression of TNF-?,TGF-? and IL-10,improve the antioxidant system,induce the level of ALT and AST.
