| Research background and purpose Since the first discovery of Helicobacter pylori(Hp)in the 1980 s,it has received extensive attention from biologists, pathologists, clinicians and other people around the world.Hp was originally a bacteria cultured in vitro by Barry J.Marshall and J.Robin Warren. When it settles in the gastric mucosa,it will destroy the protective barrier of the mucous membrane, which can not only cause gastric mucosal inflammation, erosion, ulcer and other diseases, but also closely related to the formation of inflammatory hyperplastic gastric polyps.Although the integrity of Hp infection destroying the gastric mucosa is inseparable from the inflammatory reaction that triggers the gastric mucosa, the inflammatory response is only one of the pathogenic processes.Studies have shown that the destruction of gastric mucosa is also related to various inflammatory cytokines such as various interleukins(IL)induced by Hp infection,Because they can recruit inflammatory cells to specific parts of the gastric mucosa, and these inflammatory cytokines may also have a certain correlation with the formation of inflammatory hyperplastic gastric polyps..In this paper,we mainly determined the expression levels of interleukin-1,6,8(IL-1,6,8)in normal gastric mucosa,Hp positive without gastric polyps,Hp positive with inflammatory hyperplastic gastric polyps, and explored its clinical significance to further understand Hp infection, and the relationship among the expression levels of IL-1,IL-6,IL-8,and the possible process of the formation of inflammatory hyperplastic gastric polyps caused by Hp infection. Methods 1.Participants:Selected from the patients admitted to the First Affiliated Hospital of Henan University from February 2017 to February 2019.31 cases of group A were selected from both the urease test and the 13 C breath test under the gastroscope showed Hp(-)and normal gastric mucosa without gastric polyps and other gastric mucosal lesions;30 cases of group B were selected from both the urease test and the 13 C breath test under the gastroscope showed Hp(+)and normal gastric mucosa without gastric polyps and other gastric mucosal lesions.33 cases of group C were selected from both the urease test and the 13C breath test under the gastroscope showed Hp(+)and only with inflammatory proliferative gastric polyps.2.Data collection: All patients underwent a 13 C breath test before gastroscopy, and then the gastroscopic examination was performed by the same physician and assistant. Each patient underwent a gastric mucosal tissue in the gastric curvature, small curvature of the stomach, anterior wall and posterior wall within 5 cm of the antral pylorus. The gastric mucosal tissue on the large curved side of the stomach was used for urease test.Only when the DOB value of the 13 C breath test was >4.0 and the urease test was positive, the Hp infection was counted;the remaining part of the gastric mucosa was determined by enzyme-linked immunosorbent assay(ELLSA)sandwich method to determine the interleukin-1,6 8,level.3.Statistical methods: Data was compiled using the SPSS 22.0 statistical software package. All variables were expressed as mean ± standard deviation.Statistical differences in IL-1,IL-6,and IL-8 expression levels in the three groups were analyzed by one way ANOVA using LSD test to analyze the difference between the two groups,P < 0.05 believes that the difference is statistically significant.Correlation among the expression levels of IL-1,IL6,IL-8 in the group was analyzed by spearman.Results(1).The expression levels of IL-1 in the group A,group B and group C were compared respectively. It was found that IL-1 was higher than one of the group A and group B,and the difference was statistically significant(P<0.05).IL-1 The expression level in group C was higher than that in group B,and the difference was statistically significant(P < 0.05).(2).The expression levels of IL-6 in the group A,group B and group C were compared respectively. It was found that IL-6 was higher than one of the group A and group B,and the difference was statistically significant(P<0.05).IL-6 The expression level in group C was higher than that in group B,and the difference was statistically significant(P < 0.05).(3).The expression levels of IL-8 in the group A,group B and group C were compared respectively. It was found that IL-8 was higher than one of the group A and group B,and the difference was statistically significant(P<0.05).IL-8 The expression level in group C was higher than that in group B,and the difference was statistically significant(P < 0.05).(4).The correlation analysis among IL-1,6,and 8 in group A were all P>0.05.The correlation coefficient of IL-1 and IL-8 in group B was r=0.514,P=0.004,and the correlation analysis results of IL-6 and IL-1,IL-8 were all P>0.05;the correlation coefficient of IL-1 and IL-8 in group C was r=0.527,P=0.002,and the correlation analysis results of IL-6 and IL-1,IL-8 were all P>0.05.Conclusion The expression levels of interleukin-1,6,and 8 in gastric mucosa caused by Hp infection were higher than those in normal gastric mucosa,P<0.05,with statistical difference,it may be that Hp colonizes the gastric mucosa and promotes the increase of IL-1,6,8 in expression levels. The expression levels of IL-1,6,8 in Hp-infected Inflammatory hyperplasia gastric polyps were higher than those in Hp infection without gastric polyps,P < 0.05,with statistical difference,it may be that the increase of IL-1,6,8 is conducive to the formation of inflammatory proliferative gastric polyps, this conclusion has yet to be confirmed by further research;in addition,Correlation analysis among the expression levels of IL-1,6,8 indicates that Hp infection promotes positive correlation between the expression levels of IL-1 and IL-8 in gastric mucosa. |
