Influence Of Nage Tongmai Decoction On The Expression Of MMP-9 And IL-6 In Rats With Myocardial Ischemia

Objective: The rat model of myocardial ischemia was reproduced in this experiment.Observing the pathological changes of myocardial tissue and the expression of serum MMP-9 and IL-6 in rats with myocardial ischemia induced by Nage Tongmai Decoction(NG).To explore the mechanism of this prescription in the prevention and treatment of coronary heart disease(CHD).Methods:Ninety healthy Wistar rats were randomly divided into six groups after one week of adaptive feeding,they are blank group,model group,xintong oral liquid positive control group,and Nage Tongmai decoction high dose,medium dose and low dose groups,there are 15 rats in each group.Except the blank group was fed by normal diet,rats in the other groups were fed with high-fat diet for 8 weeks.4 weeks later,the blank group and model group were given distilled water into stomach,xintong oral liquid positive control group was given Xintong Oral Liquid into stomach,and the high,middle and low dose groups were given Nage Tongmai Decoction into stomach,these intragastric administration continue for 4 weeks.Until the last 3 days of the 8th week,rats in the blank group were intraperitoneally injected with normal saline,while rats in the other groups were intraperitoneally injected with isoproterenol(0.5mg/100 g body weight),once a day until the time of getting samples.After the last intraperitoneal injection,we anesthetized rats and examined their electrocardiogram.During the treatment,we observed he general state of rats.At the end of the experiment,abdominal aorta blood was collected,and the levels of MMP-9and IL-6 in serum were measured by IHC technology.Each group rats myocardial pathological morphology was observed under the light microscope.Results:1.Pathological sections of rats in each group showed that the myocardial cells in the blank group were normal in shape and orderly arranged,and the nuclei could be uniformly stained without myocardial fibers breaking.In model group,myocardial cells were disordered,cytoplasmic congestion,edema,light stained,myocardial fibers were broken,and the structure of myocardial tissue was seriously disordered,which was significantly different from that in blank group.The pathological changes of myocardial tissue in the other groups were similar,but the degree was much less than that in the model group.The pathological changes of myocardial tissue in the high dose group of Nage Tongmai Decoction were the lightest.2.The results of serum MMP-9 and IL-6 of rats in each group: Compared with the blank group,the expression of serum MMP-9 and IL-6 in model group was significantly increased and the difference has statistical significance(P < 0.01);Compared with model group,the expression levels of serum MMP-9 and IL-6 in each dose group of Nage Tongmai Decoction and Xintong Oral Liquid group were significantly decreased and the difference had statistical significance(P < 0.05).The expression of MMP-9 and IL-6 in serum of Nage Tongmai Decoction high dose group was significantly lower than Xintong Oral Liquid group,that was a very significant difference(P < 0.01),the expression in middle dose group was significantly lower and it has significant difference(P < 0.05),however the expression has no significant difference in low dose group.Conclusion:1.Nage Tongmai Decoction can alleviate the pathological damage of myocardial cells in rats with myocardial ischemia.2.Nage Tongmai Decoction can reduce the levels of MMP-9 and IL-6 in myocardial ischemia model rats effectively,inhibit myocardial inflammation,and protect vascular endothelial cells effectively,and then improve myocardial ischemia.
