Preliminary Study On Cytokines Related To Diabetic Retinopathy

Objective:To observe the concentration changes of leukocyte cell-derived chemotaxin-2(LECT-2),interleukin(IL)-1?,soluble Inteleukin-2 receptor(sIL-2R),IL-6,IL-8,IL-10,IL-17,IL-27,IL-35,tumor necrosis factor-?(TNF-?)in the serum of patients with diabetic retinopathy(DR).Analysis of the difference in the levels of the cytokines in the serum of DR patients,and to explore its clinical significance.Method:A total of 87 subjects were enrolled,including 49 patients with proliferative diabetic retinopathy(PDR),15 patients with non-proliferative diabetic retinopathy(NPDR)and 23 normal controls.The levels of serum LECT-2,IL-1?,sIL-2R,IL-6,IL-8,IL-10,IL-17,IL-27,IL-35,TNF-? in peripheral blood were detected by enzyme linked immunosorbent assay(ELISA).The biochemical indexes were also determined.The levels of cytokines were analyzed and compared among the groups.Result:There were no significant differences in gender,age,total cholesterol,diastolic blood pressure,systolic blood pressure,triglyceride and other indicators(P>0.05).There were significant differences in the duration of diabetes and glycosylated serum protein(P<0.001;P =0.007).Serum sIL-2R levels in PDR patients were significantly higher than those in NPDR patients and normal controls(P=0.018;P<0.001).There was no significant difference between serum sIL-2R levels and normal controls in NPDR patients(P=0.262).Serum IL-6 levels in PDR patients were significantly lower than those in NPDR patients and normal controls(P<0.001,P<0.001),but there was no significant difference between the NPDR patients and the normal control group(P=0.436).Serum IL-35 levels in the PDR patients group were significantly lower than those in the normal control group(P=0.004),but there was no significant difference between the PDR patients group and the NPDR patients group,the NPDRpatients group and the normal control group(P=0.164,P=0.308).Serum LECT-2levels were significantly higher in the PDR patients than in the normal control group(P=0.014),and there was no significant difference in serum LECT-2 levels between the NPDR patients(P=0.399;P=0.251).Serum IL-1 ?,IL-8,IL-10,IL-17,IL-27,TNF-? and other cytokine levels were not significantly different between NPDR patients,PDR patients and normal controls,P>0.05.Logistic regression analysis showed that sIL-2R and IL-6 were independently associated with DR(P<0.05),and IL-35,LECT-2 and DR were not significantly independent(P>0.05).Conclusion:Serum levels of sIL-2R and LECT-2 were significantly increased in the PDR group,while serum IL-6 and IL-35 levels were significantly decreased.sIL-2R and IL-6 were independent risk factors for DR.
