Objective To observe the effects of Guweikang Granule on white blood cell count,mononuclear cell count,CD34 ~+ cell content and the expression of growth factor of VEGF,TGF-?and PDGF in peripheral blood of rats,this paper explores whether Guweikang Granule can participate in and promote the mobilization of rat bone marrow stem cells and stimulate the expression of growth factor of peripheral blood cells,so as to mobilize stem cells and increase the production of peripheral blood cells for the next step of using traditional Chinese medicine.Long factor to achieve non-invasive treatment of bone ischemic necrosis,bone defect,bone nonunion and other orthopedic diseases as a preliminary basic research.Methods Sixty SD rats were randomly divided into four groups with 15 rats in each group.Normal saline group: intragastric administration of normal saline and subcutaneous injection of normal saline;Guweikang granule group: Guweikang granule diluent,while subcutaneous injection of normal saline;G-CSF group(G-CSF group): normal saline,while subcutaneous injection of G-CSF;Guweikang granule + G-CSF(combined drug group)Guweikang granule diluent,the same.G-CSF was injected subcutaneously for 6days.Peripheral blood was taken from rats in each group for counting white blood cells and single nucleus cells.The number and percentage of CD34 ~+ cells were detected by flow cytometry,and the contents of VEGF?TGF-??PDGF in peripheral blood were detected by ELISA.Results Five days later,the peripheral white blood cells in saline group,Guweikang Granule group,G-CSF group and Guweikang Granule+G-CSF group were(7.47±0.40)×10~9/L,(7.76±0.41)×10~9/L,(19.81±0.66)×10~9/L,(20.01±0.58)×10~9/L,and the white blood cell count in Guweikang Granule group was compared with that in saline group(P>0.01).The white blood cell count in G-CSF group was higher than that in Guweikang Granule+G-CSF group.The peripheral blood leukocyte count in G-CSF group and Guweikang Granule+G-CSF group was higher than that in saline group(P<0.01).The peripheral blood mononuclear cells in saline group,Guweikang Granule group,G-CSF group and combined drug group were(3.32±0.30)×10~9/L,(4.93±0.51)×10~9/L,(10.99±0.62)×10~9/L,(12.64±0.69)×10~9/L,respectively.The number of mononuclear cells in Guweikang granule group was higher than that in saline group(P < 0.01),the number of mononuclear cells in G-CSF group was higher than that in Guweikang granule group(P < 0.01),and that in combination group was higher than that in G-CSF group(P < 0.01);the percentage of CD34 ~+ cells in Guweikang granule group,G-CSF group and combination group was(0.78±0.04)%,(3.26±0.07)%,(3.90±0.08)%,higher than that in negative control group(0.20±0.02)%(P < 0.01),in which the percentage of CD34 ~+ cells in G-CSF group was higher than that in Guweikang granule group(P < 0.01),and the percentage of CD34 ~+ cells in combination group was higher than that in G-CSF group(P < 0.01).The contents of VEGF and TGF-? in Guweikang granule group,G-CSF group and combined drug group were higher than those in saline group(P < 0.01).The contents of VEGF and TGF-? in G-CSF group were higher than those in Guweikang granule group(P < 0.01).The contents of VEGF and TGF-? in combined drug group were higher than those in G-CSF group(P< 0.01).Compared with the normal saline group,the content of PDGF in Guweikang granule group and combined drug group was higher than that in saline group(P < 0.01),but there was no difference between G-CSF group and saline group(P > 0.01).Conclusion Guweikang Granule can mobilize stem cells and increase the expression of VEGF,TGF-? and PDGF in peripheral blood.The mechanism of Guweikang Granule in treating osteonecrosis may be related to this.Guweikang granule combined with G-CSF has the best effect on mobilization of stem cells,and can significantly increase the levels of VEGF,TGF-? and PDGF in peripheral blood while mobilizing stem cells.This suggests that we can try to use Chinese herbal medicine as a stem cell mobilizer or assistant stem cell mobilization in future research,which can increase the number of peripheral blood stem cells and the content of growth factor in peripheral blood,and provide sufficient cell source and growth factor conditions for the repair of damaged tissues.The experiment proved that the use of traditional Chinese medicine in the mobilization of stem cells in the treatment of orthopedic diseases in cytology is feasible,has research significance and application prospects.
