ObjectiveRapid identification of Polygonatum cyrtonema Hua from different areas and Polygonatum Mill.by three methods,to provide a scientific evidence for studying quality control and evaluation of Polygonatum sibiricum.MethodsTo identify the Polygonatum cyrtonema Hua from different areas and Polygonatum Mill.by electrochemical fingerprint technology,infrared spectrum technology and Fourier-Transform Infrared Microspectroscopy.ResultsThe electrochemical fingerprints of Polygonatum sibiricum were studied,the optimum conditions of the B-Z oscillation were as follows,the temperature of 310 K,adding 0.1000 g of sample powder and the speed of 500 r/min.The electrochemical fingerprints and Characteristic Parameters of Polygonatum cyrtonema Hua from different origin and Polygonatum Mill.were obtained and discussed.The result of Fourier transform infrared spectroscopy showed that the main absorption peaks of Polygonatum cyrtonema Hua were in the vicinity at 3270?2933?1623?1411?1319?1269?1103?1018?926?865?815 cm-1.The FT-IR spectra of Polygonatum cyrtonema Hua from different areas were similar,but the intensity and position of absorption peaks were different.The absorption peak near 1018 cm-1 is the characteristic absorption peak of glucose.From the infrared spectrum,it can be inferred that the content of glucose components in Chizhou,Yuexi,and Yichun is high.Huanggang,Huangshan,Shitai are less.Xi'an County,Xiuning,Jinzhai County are relatively the least.The infrared spectra of six species of Polygonatum Mill.are different in the position,shape and intensity of absorption peaks,so it can be inferred that there are obvious differences in chemical composition and content among different plants of Polygonatum Mill..The second derivative spectrum can spectrally strip the overlapping peaks in the original spectrum,further improving the resolution of the spectrum.Compared with the ordinary infrared spectra spectrum,the second derivative infrared spectrum further intuitively verify the above results.The FTIR images of Polygonatum Mill.were studied.The optimum experimental conditions were as follows:Detection mode of transmission,BaF2 window and 30?m slice thickness of sample.The FTIR images of six species of Polygonatum Mill.were quite different.Three types of vascular bundles were found in rootstocks of Polygonatum Mill.by observing microscopic images.The vascular bundles of Polygonatum cyrtonema Hua and Polygonatum sibiricum Red.are incomplete amphivasal vascular bundle.The vascular bundles of Polygonatum kingianum Coll.et Hemsl and Polygonatum verticillatum?L.?Al are mostly amphivasal vascular bundle,and a few are collateral bundle.The vascular bundles of Polygonatum filipes Merr.are collateral bundle and incomplete amphivasal vascular bundle.The vascular bundles of Polygonatum zanlanscianense Pamp.are amphivasal vascular bundle and incomplete amphivasal vascular bundle.Observation of chemical imaging revealed that the contents of polysaccharides in Polygonatum Mill.ranged from high to low as follows:Polygonatum cyrtonema Hua>Polygonatum zanlanscianense Pamp.?Polygonatum kingianum Coll.et Hemsl>Polygonatum filipes Merr.?Polygonatum sibiricum Red.>Polygonatum verticillatum?L.?Al.ConclusionThe electrochemical fingerprints of Polygonatum cyrtonema Hua from different origins and Polygonatum Mill.showed significantly different characteristics.Rapid identification Polygonatum cyrtonema Hua from different origins,six species of Polygonatum Mill.through the electrochemical fingerprints and the information parameters.A rapid,simple and economical method was developed for identification of geographical origin and evaluation of Polygonatum sibiricum samples by the electrochemical fingerprints.Identification of Radix Polygonatum sibiricum by infrared spectroscopy combined with second derivative spectroscopy,not only provide the information of main chemical constituents in Polygonatum sibiricum,but also classify and identify Polygonatum cyrtonema Hua from different areas and six species of Polygonatum Mill..The FTIR method was proved to be feasible for identify rapidly Polygonatum cyrtonema Hua from different areas and Polygonatum Mill..The chemical images of Polygonatum Mill.were quite different.Microscopic infrared imaging technology combines microscopic imaging with functional group chemical analysis of infrared spectroscopy.It can not only image the transverse section of rootstock of Polygonatum Mill.,but also provide spectral information of each point in the transverse section space,observe the distribution of its effective components,and carry out semi-quantitative research.The method has advantages such as intuitive,accurate and high sensitive,it comprehensively analyzes the differences in the intrinsic quality of Polygonatum Mill..
