| Objective:Through the detection of CD8~+CD28~+T lymphocytes content in the serum and liver tissues of patients with hepatitis B at different infection stages,observe the expression of CD8~+CD28~+T lymphocytes in different groups and discuss the relationship between CD8~+CD28~+T lymphocytes and peripheral blood HBs Ag,and explore its clinical significance.Methods:A total of 80 patients with hepatitis B and 20 healthy individuals were recruited in 902 nd hospital of PLA between April 2018 and January 2019,including patients in immune tolerance phase(n=20),immune clearance phase(n=20),low replication phase(n=20),and re-activation phase(n=20).The percentages of CD8~+CD28~+T lymphocytes in peripheral blood were measured by flow cytometry,and which in tissues were measured by Immunohistochemistry.The experimental data were collected for statistical analysis.Results:(1)With the results of flow cytometry and Immunohistochemistry,there were significantly differences among those groups(P < 0.05),the percentages of CD8~+CD28~+T lymphocytes in serum among healthy persons,immune tolerance phase,immune clearance phase,low replication phase,re-activation phase were(26.2±3.5)%,(26.6±3.5)%,(40.3±5.5)%,(20.0±4.4)%,(22.3±2.0)%,while the percentages of CD8~+CD28~+T lymphocytes in tissues among immune tolerance phase,immune clearance phase,low replication phase,re-activation phase were(13.1±3.5)/HP,(31.1±5.3)/HP,(6.2±2.4)/HP,(8.7±1.9)/HP.(2)There were significant differences among low serum HBs Ag level group,moderate serum HBs Ag level group and high serum HBs Ag level in serum as well as tissues(P < 0.05).The CD8~+CD28~+T lymphocytes in serum with low,moderate and high serum HBs Ag level were(22.7±6.3)%,(27.0±8.1)%,(32.7±8.6)% respectively,while were(9.4±7.8)%,(14.3±9.8)%,(21.2±10.4)% in liver tissues respectively.Conclusion:(1)There are significant differences in the percentages of CD8~+CD28~+T lymphocytes in patients with hepatitis B at different immune phases,which might be related to the dysfunctions of immune system activated by the viral infection.(2)HBs Ag level may reflect the activation of liver immune response. |
