Objective: To explore the expression of E-cadherin(E-cadherin),beta-catenin(beta-cat),matrix metalloproteinase-9(MMP-9)in ectopic endometrial tissue,eutopic endometrial tissue and normal endometrial tissue of patients with endometriosis(EMS),by detecting the expression of E-cadherin(E-cadherin),beta-catenin(beta-cat),matrix metalloproteinase-9(MMP-9).The significance of expression of beta-catenin,MMP-9 in ectopic endometrium,eutopic endometrium and normal endometrium of non-EMS patients was explored.The relationship among E-cadherin,beta-catenin and MMP-9 was explored,and their influence on the occurrence and development of endometriosis was also explored,and the pathogenesis of endometriosis was explored.Methods: 1.46 cases of ectopic endometrial tissues in EMS patients were selected,which were clearly diagnosed by operation and pathology after operation(all of them had clean menstruation for 3-7 days and were endometrial tissues in proliferative stage).At the same time,46 cases of eutopic endometrial tissues in the same patient were obtained.Endometrial tissue(including eutopic endometrium and ectopic endometrium)from EMS patients was used as experimental group.The endometrial tissues of 40 patients with uterine fibroids or cervical high-grade intraepithelial neoplasia who underwent hysterectomy were selected as the control group.2.The expressions of E-cadherin,beta-catenin and MMP-9 in ectopic endometrium,eutopic endometrium and normal endometrium were detected by immunohistochemical staining(Elivision)and compared.To explore the relationship between the three factors and the occurrence and development of EMS patients.3.SPSS20.0 was used to process the data.The results were analyzed with clinical data.C2 test was used between groups.Spearman grade correlation analysis and Fisher exact probability analysis were used between E-cad,beta-cat and MMP-9.Statistical results showed that there was significant difference between P < 0.05 and P < 0.05 by C2 test,and Spearman grade correlation analysis showed significant correlation between P < 0.05 and P < 0.05.Results: 1.E-cad was mainly expressed in the endometrium of endometrial glandular epithelial cells.The positive rates of E-cad in the abnormal endometrium,eutopic endometrium and normal endometrium of EMS were 52.17%,78.26% and 100%,respectively.The expression of beta-catenin was mainly in the endometrium and cytoplasm of endometrial glandular epithelial cells,which was the cytoskeleton protein and intracellular signal transduction molecule.The positive rates of abnormal expression of beta-cat in abnormal endometrium,eutopic endometrium and normal endometrium tissues of EMS were 84.78%,60.86% and 32.50%,respectively.MMP-9was an extracellular matrix catalase,mainly expressed in endometrial glandular epithelium and cytoplasm.The positive rates of abnormal expression of MMP-9 in abnormal endometrium,eutopic endometrium and normal endometrium tissues of EMS were 84.78%,60.86% and 32.50%respectively.They were 82.61%,56.52% and 27.50% respectively.2.Comparing the expression levels of E-cad,beta-cat and MMP-9 in three groups of endometrium,the expression level of E-cad in ectopic endometrium of EMS was lower than that in eutopic endometrium,and that in eutopic endometrium was lower than that in normal endometrium.The expression level of E-cad in eutopic endometrium and ectopic endometrium decreased significantly in EMS patients(p < 0.05 and P < 0.01).The expression level of E-cad in eutopic endometrium tissue of EMS patients was significantly different from that in ectopic endometrium tissue(p < 0.05).The expression level of E-cad in eutopic endometrium tissue of EMS patients was significantly different from that in normal endometrium tissue of non-EMS patients(p < 0.05).The expression level of E-cad in ectopic endometrium tissue of EMS patients was significantly different from that of non-EMS patients(p < 0.05).The expression level of beta-cat in ectopic endometrium tissue of EMS patients was higher than that in eutopic endometrium and normal endometrium tissue of non-EMS patients.The expression intensity of beta-cat in eutopic endometrium tissue of EMS patients was higher than that of non-EMS patients.There was no significant difference in the expression of beta-cat between eutopic and ectopic endometrium in EMS patients(p < 0.05).There was no significant difference in the expression of beta-cat between eutopic endometrium in EMS patients and normal endometrium in non-EMS patients(p > 0.05).There was no significant difference in the expression of beta-cat between eutopic endometrium and ectopic endometrium in EMS patients(p > 0.05).The expression level of MMP-9 in ectopic endometrium tissues was significantly higher than that in eutopic endometrium tissues,in eutopic endometrium tissues was higher than that in normal endometrium tissues,and in eutopic endometrium tissues and ectopic endometrium tissues of EMS patients,the expression level of MMP-9 was significantly higher than that in eutopic endometrium tissues and ectopic endometrium tissues.The expression level of MMP-9 in eutopic endometrium tissue of EMS patients was significantly different from that in normal endometrium tissue of non-EMS patients(p < 0.05).The expression level of MMP-9 in ectopic endometrium tissue of EMS patients was significantly different from that in normal endometrium tissue of non-EMS patients(p < 0.05).The expression level of MMP-9 in ectopic endometrium tissue of EMS patients was significantly different from that in normal endometrium tissue of non-EMS patients.The difference was significant(p < 0.01).3.Abnormal expression of E-cad protein and beta-cat protein in ectopic endometrium tissues of EMS patients indicated that there was a significant negative correlation between E-cad protein and beta-cat protein(rs=-0.428,P=0.003);abnormal expression of E-cad protein and MMP-9 protein was significantly and moderately negatively correlated(rs=-0.439,P=0.002);abnormal expression of beta-cat protein and MMP-9 protein was significantly and moderately positively correlated(rs=0.604,P=0.000).Conclusion: Abnormal expressions of E-cad,beta-cat and MMP-9 are found in EMS.The low expression of E-cad in EMS and the high expression of beta-cat and MMP-9 in EMS have a great influence on the occurrence and development of EMS,which is one of the reasons for the stronger adhesion and invasion of ectopic endometrium and eutopic endometrium in EMS patients.The low expression of E-cad in EMS and the high expression of beta-cat and MMP-9 in EMS promote the occurrence and development of EMS.Therefore,the simultaneous detection and comparative analysis of the above three indicators are of great significance in exploring the pathogenesis of endometriosis.The combined monitoring of E-cad,beta-cat and MMP-9 in EMS can also be used as an important means to predict the effect of EMS treatment and the long-term development of EMS.
