ObjectiveTo study the expression of E-cad,a'-cat and FAK in normal endometrium^ ectopic and eutopic endometrium from woman with endometriosis, thereby to determine the role of E-cad,a -cat and FAK and their relationship in the pathogenesis of endometriosis.Method1.46 specimans with endometriosis (22 cases of proliferative phase and secretory phase in 24 cases) meanwhile 46 eutopic endometrium cases were selected as research group,42 cases of normal endometrium, of infertility patient because of tubal factor (20 cases of proliferative phase and secretory phase 22 cases) were selected as a control group. the expression of E-cad,a -cat and FAK were detected in eutopic and ectopic endometrium of endometriosis as well as normal endometrium by means of Immunohistochemistry SP method, to compare the expression of E-cad,a-cat and FAK among the three groups.2. On the basis of what we saw in the laparoscope or laparotomy,we class-ified the 46 endometriosis patients into two groups(Ⅰ~Ⅱstage as A group,Ⅲ-Ⅳstage as B group),according to the classification of r-AFS in 1985,to compare the expression of E-cad,a-cat and FAK between the two groups.3. The result was analyzed by SPSS13.0 statistical software.Results1①E-cad and a-cat were mainly expressed in the membrane and cytoplasm of glandular epithelial cells in normal endometrium, FAK was located in normal endometrial tissue in the cytoplasm of gland cells and stromal cells. The positive degree of staining of E-cad,a-cat and FAK among the three groups were compared,the difference was very significant (P<0.01) in each two groups. ②The expression of E-cad in ectopic endometrium was lower than in eutopic endometrium, its expression in eutopic endometrium was lower than in normal endometrium.Its expression at secretory phase was higher than at proliferative phase, which was no statistically significant in the same group(P> 0.05). Compared with the control group at proliferative phase, the expression of E-cad in eutopic / ectopic endometrium from patients with endometriosis was significantly decreased at the same phase (P<0.05 & P<0.01),the same was true at secretory phase.The difference expression of E-cad in eutopic / ectopic endometrium was no significant at proliferative phase,while at secretory phase it was significant.③The expression of a-cat in ectopic endometrium was lower than in normal/eutopic endometrium, its expression in eutopic endometrium was lower than in normal endometrium,the difference in each two groups at the same phase was significant.Its expression at proliferative phase was higher than at secretory phase, which was no statistically significant in the same group(P> 0.05).④The expression of FAK in ectopic endometrium was higher than in eutopic endometrium,its expression in eutopic endometrium was higher than in normal endometrium,the difference was statistically significant.Its expression at secretory phase was higher than at proliferative phase, there was no significant difference between the two phases in the same group (P> 0.05). The expression of FAK was no significant difference at proliferative phase among the there groups(P>0.05),while at secretory phase compared with the control group, the expression of FAK in eutopic/ectopic endometrium was significantly increased at the same phase (P<0.05 &P<0.01).2 The positive degree of staining of E-cad and a-cat in group B(stageⅢ~Ⅳ) was lower than in group A (stageⅠ-Ⅱ), The positive degree of staining of FAK in group B (stageⅢ-Ⅳ) was higher than in group A (stageⅠ-Ⅱ). There was significant difference between the two groups. (P<0.01)3 The expression of E-cad and a-cat in ectopic endometrium of endometriosis were positive correlation.4 The expression of E-cad and FAK in ectopic endometrium of endome-triosis were negative correlation. 5 The expression of a-cat and FAK in ectopic endometrium of endomet-riosis was negative correlation.Conclusion1 The decreased expression of E-cad,a-cat and the increased expression of FAK were involved in the invasion of endometriosis and they were one of the reasons why ectopic and eutopic endometrial cells have stronger invasive ability than that of normal endometrial cells.2 The different expression level of E-cad, a-cat and FAK had something to do with clinical phase of endometriosis.3 The expression of FAK may inhibit the expression of E-cad and a-cat in ectopic endometrium, The high expression of FAK co-promoted the progress of endometriosis with the low expression of E-cad and a-cat.
