Objective:To investigate the effect of the adjuvant therapy of different osmotic pressure of enteral nutrition(EN)solution on intestinal immune barrier,hemodynamics,inflammatory factors and free radical metabolism of ulcerative colitis(UC)rats.Methods:UC rats were prepared according to the literature,which divided into the following groups:UC model group,200 mmol/L EN group,300 mmol/L EN group,400 mmol/L EN group,500 mmol/L and 600 mmol/L EN group.And the normal health rats were collected as the control group.Each group contains 10 rats,with five males and five females.All UC rats were fed with feeding tube by gastrostomy.Each UC groups were given 200mmol/L-600 mmol/L EN for 7 days,respectively.UC model group and the control group were fed with equal volume of 5%glucose solution for 7 days.The survival state and body weight of all the rats were observed,DAI was calculated,and the intestinal barrier function was examined by lactulose/mannitol method.At the end of the EN treatment,the cases of microvascular branches,the diameter of the jejunum mesenteric membrane,and the blood flow state scores were recorded under anesthesia.Each rat's abdominal aorta blood was collected and the content of plasma endotoxin was determined by azo reagent matrix azo color.The contents of TNF-a,IFN-y,IL-6 and IL-2 in plasma were determined by ELISA.HE staining was employed to observe the damage of colonic mucosa in colon tissue,CMDI was evaluated,and the expression levels of MAdCAM-1(Mucosal Addressin cell adhesion molecule-1),CD4 and CD8 were detected by immunohistochemistry.The contents of SOD,MDA,MPO and NO in colon tissue were measured separately.Results:UC rats were apathetic,and their activity was significantly weakened,the whole body lost weight significantly and became thinner,the stool was soft,the blood was loose,the hair was loose and dry,the DAI increased,and the intestinal barrier function decreased.After 7 days' EN treatment,the general health condition of the rats were improved,the DAI data decreased.The state of the 300 mmol/L EN group showed the best result.On the 3rd day of EN treatment,the intestinal barrier function in the 300mmol/L?600mmol/L groups were significantly improved.Among them,300mmol/L EN group was the lowest in lactulose/mannitol.The cases of vascular branches,tube diameter and fluidity of the mesenteric microvessels in the UC model groups were significantly lower than those in the control group.These indexes were significantly higher in the EN treatment groups than in the UC model group.In the UC group,the colonic tissue showed inflammatory changes,the cases of crypts was decreased with damage,with intestinal epithelial goblet cell loss and destroyed,and a large amount of inflammatory cells(monocytes,neutrophils and eosinophils)infiltrated.In the EN treatment groups,the morphology of colon tissue in rats were improved to some extent compared with the UC model group.Compared with control group,the expression of MAdCAM-1 and CD4 in colon tissue were significantly:increased,CD8 expression was significantly decreased,MDA,MPO and NO content were significantly increased,while SOD content was significantly decreased.In the UC model group,the endotoxin and inflammatory factors TNF-a,IFN-y and IL-6 had increased significantly,while the inhibitory factor IL-2 decreased.These indicators were improved in each group with the treatment of EN.The 300mmol/L?600mmol/L group were significantly different from the UC model group,and which showed a dose-dependent dose manner,the indicators showed the 300mmol/L EN group performed the best compared to other groups.Conclusion:EN can improve the intestinal barrier,hemodynamics,inflammatory factors and free radical metabolism of UC.Especially the intestinal nutrition with osmotic pressure of 300mmol/L has obvious advantages in improving the intestinal barrier,hemodynamics,inflammatory factors and free radical metabolism of UC.
