| Objective:This study was to detect the expression level and clinicopathological significance of EGFR,Rac1 and Pak1 proteins in gastric cancer tissues and normal tissues,and provided experimental basis for molecular targeted diagnosis and treatment of clinical gastric cancer.Methods:The paraffin pathological tissues of patients with well-preserved gastric cancer after surgery in our hospital from 2011 to2017 were used as experimental subjects,and the expression levels of EGFR,Rac1 and Pak1 in gastric cancer,adjacent normal tissues and normal tissues were detected by immunohistochemical SP method,then the SPSS system was used to analyze its relationship with clinical pathology.Results:1.EGFR protein is mainly located on the cell membrane and nuclear membrane.There are different expressions in gastric cancer,cancer collateral and normal tissues,and the expressions of the three were significantly different(x~2=24.458,P<0.05),the expression of EGFR protein in para-cancer and normal tissues was significantly lower than that in gastric cancer tissues(P<0.05).Moreover,EGFR expression was different in gastric cancer with different degrees of differentiation,TNM stage and lymph node metastasis,the EGFR expression positive rate in the poorly differentiation and high differentiation groups of gastric cancer were 87.2%and 74.8%,there were significant differences between the two groups(x~2=4.674,P<0.05).The positive rate of EGFR expression inⅠ+ⅡandⅢ+Ⅳwas 69.4%and 83.0%,and there are significant differences between them(x~2=5.053,P<0.05).Lymph node metastasis and no lymph node metastasis were 82.8%and 70.3%,and there are significant differences between them(x~2=4.303).2.Rac1 is mainly located in the cytoplasm or membrane.There are different expressions in gastric cancer,cancer collateral and normal tissues,and the expressions of the three were significantly different(x~2=28.159,P<0.05),the positive rate of Rac1 protein expression in cancer and normal tissues was significantly lower than that in gastric cancer tissues(P<0.05).Moreover,Rac1 expression was different in gastric cancer with different degrees of differentiation,TNM stage and lymph node metastasis,the Rac1 expression positive rate in the low differentiation and high differentiation groups of gastric cancer were84.6%and 70.6%,there were significant differences between the two groups(x~2=5.347,P<0.05).The positive rate of Rac1 expression inⅠ+ⅡandⅢ+Ⅳwas 61.3%and 81.1%,and there are significant differences between them(x~2=9.511,P<0.05).Lymph node metastasis and no lymph node metastasis were 80.3%and 64.1%,and there are significant differences between them(x~2=6.456,P<0.05).3.Pak1 is mainly located in the cytoplasm or membrane.There are different expressions in gastric cancer,cancer collateral and normal tissues,and the expressions of the three were significantly different(x~2=24.964,P<0.05),the positive rate of Pak1 protein expression in cancer and normal tissues was significantly lower than that in gastric cancer tissues(P<0.05).Moreover,Pak1 expression was different in gastric cancer with different degrees of differentiation,TNM stage and lymph node metastasis,the Rac1 expression positive rate in the low differentiation and high differentiation groups of gastric cancer were83.3%and 69.9%,there were significant differences between the two groups(x~2=4.479,P<0.05).The positive rate of Rac1 expression inⅠ+ⅡandⅢ+Ⅳwas 64.5%and 78.6%,and there are significant differences between them(x~2=4.688,P<0.05).Lymph node metastasis and no lymph node metastasis were 78.3%and 65.6%,and there are significant differences between them(x~2=3.888,P<0.05).4.The positive rate of Rac1 in EGFR positive group(82.86%)was higher than that in EGFR negative(47.83%),the difference was significant(P<0.05),and coefficient of correlation r=0.404.The positive rate of Pak1 in EGFR positive group(81.14%)was higher than that in EGFR negative(50.00%),the difference was significant(P<0.05),and coefficient of correlation r=0.452.The positive rate of Pak1 in Rac1positive group(82.04%)was higher than that in Rac1 negative(51.85%),the difference was significant(P<0.05),and coefficient of correlation r=0.430.Conclusion:1.The high expression of EGFR,Rac1 and Pak1 in gastric cancer is closely related to the degree of differentiation,TNM stage and lymph node metastasis of gastric cancer.2.The expressions of EGFR,Rac1 and Pak1 in gastric cancer are positively correlated with each other. |
