fried before used" is a summary to processing methods of semen Chinese medicine.Semen Chinese medicine include some TCM of which the medicinal parts are fruits or seeds.It is traditionally thought that fried semen Chinese medicine held better therapeutical effects on diseases.Luo Zhouyan,a famous doctor in dynasty Ming,said that semen Chinese medicine must be fried and grinded before used in case the properties were not out.Then,what is the scientific connotation of this word?At present,the studies on this processing mechanism are mostly focused on the changes of chemicals’ contents or pharmacological actions before and after processing.The experiments on pharmacokinetics of semen Chinese medicine were seldomly searched in the database like Pubmed.Method of multi-components pharmacokinetics is always used to illustrate the mechanism of absorption,distributionmetabolism and excretion when the medicine was given into the bodies.This method was used to study the effects of processing on pharmacokinetics of four semen Chinese medicine.In the preliminary stage,chemicals’ separation and identification of four Chinese medicine Viticis Fructus,Vaccariae Semen,Descurainiae Semen and Astragali Complanati Semen were finished.At the same time,the changes of chemical compounds were studied.Based on these,components in plasma were chosen and quantitatively analyzed.Then,pharmacokinetics of these components in raw and processed TCM were studied.The results help to illuminate the pharmacodynamic material basis and explain the processing mechanism "All semen Chinese medicine must be fried before used".This paper is expanded mainly from five parts.First,a comprehensive review on the progress and research methods of processing mechanism and the processing mechanism "All semen Chinese medicine must be fried before used" was completed through searching and reading literature from databases like CNKI.Wanfang Med Online,Cqvip,Pubmed or books about TCM processing.Processing mechanisms of most TCM were not clear but several TCM which were poisonous or obviously effective.The research methods were mainly from two aspects chemicals’ analysis and pharmacology.In the aspect of chemicals’ analysis,HPLC.LC-MS and GC-MS were commonly used.Researches on the processing mechanism "All semen Chinese medicine must be fried before used" were still focused on the changes of chemicals,contents and pharmacologic actions.Seldomly the studies in vivo like pharmacokinetics were seenSecond,the effects of salt processed on pharmacokinetics of Astragali Complanati Semen were studied.Rutin as internal standard,a LC-MS/MS method for quantification of complantatoside A and B was developed,validated and used to study the pharmacokinetics of raw and salt processed Astragali Complanati Semen in rats.The Tmax value of complantatoside B in raw group were 0.37 ± 0.27 h,and the Cmax and AUC0-12 values increased significantly in processed group while MRT0-12 value was decreased.It means that processing promoted the absorption and accelerate the excretion rate of complantatoside B.Cmax and MRT0-12 values of complantatoside A in raw group were 14.72 ± 11.13 μgL-1,3.93 ± 0.26 h while 35.64 ± 21.99 μg L-1,1.43 ±0.24 h in processed group(p<0.05).It showed that processing could promote the absorption and shorten the excretion timeThird,the effects of processing on pharmacokinetics of Vaccariae Semen were researched.Taking buddleoside as internal standard,a LC-MS/MS method for quantification of vaccarin,saponarin and isovitexin-2"-O-arabinoside was developed.fully validated and applied to studies of pharmacokinetics.Relevant data was processed as described before.As a result,there were no significant difference of vaccarin and saponarin in Tmax and Cmax.But values of MRT0-24 of these two compounds were contracted from 8.85 ± 2.02 h.6.01 ± 3.06 h to 6.02 ± 1.26 h,2.84 ± 1.32 h separately.which indicated that processing made for excretion of vaccarin and saponarin.Values of Cmax and AUC0-24 of isovitexin-2"-O-arabinoside obviously increased while MRT0-24 decreased,which manifested that processing increased the degree of adsorption and the excretion time.Forth,the effects of processing on pharmacokinetics of Descurainiae Semen were observed.Complantatoside A as internal standard,a LC-MC/MS method for quantification of QGG and sinapic acid was developed,validated and applied to pharmacokinetics study of raw and processed Descurainiae Semen in rats.It showed that the values of Cmax and Tmax of QGG were 17.90±9.32 μg·L-1,2.04±1.96 h.Relevant pharmacokinetic parameters of raw and processed group showed no differences.The concentration of sinapic acid reached the top at 0.27±0.09 h,and two peaks were observed within 12 hours.Both the values of AUC0-24 and MRTO-24 were obviously increased compared with raw group,which explained that processing increased the absorption of sinapic acid and slowed down the excretion rate.Fifth,the effects of processing on pharmacokinetics of Viticis Fructus were studied.Butylparaben taken as internal standard,a simple and fast LC-MS/MS method for quantification of vitexicarpin and isoorientin in rat plasma was developed,fully validated and subsequently well used to determinate the concentrations of different plasma samples collected from rats which were given same dosage of extract of raw or processed Viticis Fructus.DAS 3.2.6 and SPSS 23.0 were used to calculate the pharmacokinetic parameters and perform for statistical analysis using T test separately.It seemed that there were no
