Part I Expression of IL-34 in patients with primary liver cancer and its clinical significanceObjective:To investigate the expression of interleukin 34(IL-34)in cancer and adjacent non-cancerous tissues from patients with primary liver cancer.Methods:The tumor tissue specimens of 93 patients(65 men and 28 women,age from 31 to 88 years old,median age was 62 years old)with primary liver cancer who underwent surgical resection in Wuxi No.4 People's Hospital from 2013 to 2018 were collected,and 74 patients(45 men and 20 women,age from 34 to 88 years old,median age was 61 years old)with the adjacent non-cancerous tissues specimens were collected,a total of 167 cases.All surgical specimens were pathological confirmed diagnosis of primary liver cancer and none patient had chemotherapy or radiotherapy before operation.Immunohistochemistry was used to detect the expression and location of IL-34 on paraffin sections in collected 167 cases,and the relationships between IL-34 expressions and clinical pathologic characteristics was analyzed.Results:The immunohistochemical staining results showed that the expression of IL-34 in cancer tissues was significantly higher than that in adjacent non-cancerous tissues.IL-34 mainly expressed in hepatocytes,localized in cytoplasm instead of cholangiocytes.The expression of IL-34 was associated with age and differentiation status,but not associated with gender,tumor size or pathological TNM stage.Conclusion:The expression level of IL-34 in cancer tissues is higher than that of adjacent non-cancerous tissues,suggesting that IL-34 is associated with the development of primary carcinoma of liver.Part ? Role of IL-34 in the development of orthotopic liver cancerObjective:To investigate the role of IL-34 in the development of orthotopic liver cancer.Methods:In order to establish orthotopic liver cancer in mice,Hepa 1/6 cells were hydrodynamicaly injected from tail vein.Meanwhile,minicircle-IL-34 or minicircle plasmids were hydrodynamicaly injected from tail vein respectively.Mice were sacrificed and the tumor numbers in liver were calculated two weeks later.Results:In orthotopic liver cancer model,the numbers of liver tumors from mice hydrodynamicaly injected with minicircle-IL-34 are significantly more than those from mice hydrodynamicaly injected with minicircle.Conclusion:IL-34 may promote the development of liver cancer.
