Objective: To study the effects of Yipi Yanggan(YPYGF)decoction on diethylnitrosamine(DEN)-induced precancerous lesions and TGF-?1and Smad3 in rats,and to explore the traditional Chinese medicine prevention and treatment for precancerous lesions.Methods: 100 male Wister rats of SPF grade were enrolled in the experiment after one week of adaptive feeding.They were randomly divided into blank group,model group,YPYGF high-dose group,Yipi Yanggan method low-dose group and Hugan tablets group.Peritoneal injection of DEN model of precancerous lesions,1 time / d,YPYGF high dose group 1.0g / ml,10 ml / kg YPYGF gavage,1 / d;YPYGF the low-dose group of liver was administered with 0.5g/ml,10ml/kg once daily,and the Hugan tablets were administered with 921mg/kg rat body weight once a day;continuous administration to 16 weeks,16 weeks The rats were sacrificed 24 hours after gavage.The general condition of the rats was observed before sacrifice.After the rats were sacrificed,blood was collected from the heart,and the liver was taken out.The liver function indexes ALT,AST,GGT,ALP and TBIL were detected for hematoxylin-eosin(The pathological changes of the liver were observed by HE staining).The degree of fibrosis was observed by Masson staining.The expression of TGF-?1 and Smad3 was detected by immunohistochemistry.Results:1.YPYGF can improve the general condition of DEN model rats with precancerous lesions.The rats in the high dose group of YPYGF are generally in good condition.The rats in the low dose group and the Hugan Tablet group have no general state.Significant differences were slightly worse than those in the high-dose group.2.YPYGF can significantly improve the liver function of DEN-induced liver cancer precancerous lesions: compared with the blank group,the ALT,AST,GGT,ALP and TBIL of the model group increased significantly.The function was significantly decreased.Compared with the model group,the ALT,AST,GGT,ALP,and TBIL of the YPYGF large and small dose group and the Hugan tablets group were significantly lower,and the liver function was improved.The YPYGF high dose group was the lower dose group.The effect of Hugan tablets group was better(P<0.05).3.In the blank group of rat liver tissue,a small amount of hepatocytes were positively expressed.TGF-?1 and Smad3 were significantly expressed in the model group,mainly in precancerous lesions and surrounding tissues,showing dark brown staining of cells as positive cells.The YPYGF large and small dose groups and Hugan tablets group can significantly inhibit the expression of TGF-?1 andSmad3,and the high dose group was the best(P<0.05).Conclusion: 1.YPYGF can significantly improve the liver function of DEN-induced liver cancer precancerous model rats,and has hepatoprotective effect;2.YPYGF has certain pathological morphology on liver cancer precancerous lesions.The effect is that the hyperplasia of the proliferating cells is significantly smaller,the binuclear cells are reduced,and the inflammation is milder,while the high-dose group is better;3.YPYGF can improve the degree of fibrosis in the precancerous lesions of liver cancer,and the high-dose group is obvious.It is better than the low-dose group and Hugan tablets group;4.YPYGF has a retarding effect on the precancerous lesions of liver cancer in rats,and its effect may be related to the inhibition of TGF-?1 and Smad3 protein expression.
