Preliminary Research On The Targeted Regulation Of Mps1 Protein Expression In 143B Cell Osteosarcoma Nude Mice Model

Objective:To investigate the effect of targeted reg?lation of Mps1 on the development of 143B cell osteosarcoma model.Methods:Twenty female nude mice were randomly divided into 4 groups:Mps1overexpression group was injected with Mps1 overexpressing 143B cells;empty virus vector group was injected with empty virus vector to transfect 143B cells;control group and Mps1inhibition group were injected with 143B cells.After the tumor formation of each group of nude mice,the inhibition group was injected intraperitoneally with Mps1 inhibitor,Mps1overexpression group,empty virus vector group and control group were injected with the same amount of normal saline.The growth of the osteosarcoma model was observed and the tumor formation was observed.The nude mice were sacrificed in the fifth week.The expression of Mps1 in the tumors of each group was detected by immunohistochemistry.The pathological changes of the tumors were observed by HE staining.Res?lts:Nude mice in each group developed tumors after(6±2)days,and the tumor formation rate was 100%.Compared with the control group,the Mps1 overexpression group had the fastest growth volume,the average volume was(2366±299)mm~3(P<0.05),and the average weight was(3.23±0.2)g(P<0.05).In the inhibition group,the tumor volume growth rate was the slowest.On the fifth week,the average volume of the tumor was(350±63)mm~3(P<0.05),and the average weight was(0.97±0.68)g(P<0.05).The res?lts of immunohistochemistry showed that Mps1 expression level was the highest in Mps1overexpression group(P<0.05),while Mps1 inhibition group had the lowest expression level(P<0.05).Conclusion:High expression level of Mps1 has a protective effect on the survival of osteosarcoma cells and promotes the development of osteosarcoma.By inhibiting Mps1kinase activity,the survival and growth ability of osteosarcoma cells is significantly reduced.
