Objectives: To study the expression and correlation of Krüppel-like factor 4 and Ki-67 protein in cholangiocarcinoma,and to explore their mechanisms in the occurrence and development of cholangiocarcinoma,so as to provide theoretical support for the treatment of cholangiocarcinoma.Methods:The complete clinical datas and pathological datas of 95 cases of paraffin samples of cholangiocarcinoma tissues from the Second Hospital of Hebei Medical University during the period from 2010.01.01 to 2018.12.30 were collected and collated.25 cases of paraffin samples of paracancerous tissues of cholangiocarcinoma proved to be negative by pathology were selected as negative control.Immunohistochemistry(S-P)was used to detect the expression of KLF4 and Ki-67 proteins in these specimens,and we combined the age,sex,tumor size,differentiation,lymph node metastasis,stage and other factors to make a comprehensive analysis.The results were analyzed by SPSS21.0 statistical software.Results:1.The positive expression rates of Krüppel-like factor 4 and Ki-67 in cholangiocarcinoma tissues were 41.0% and 73.7% respectively,which were significantly different from those in negative control group(P<0.05).2.The expression level of KLF4 in stage ?+? was significantly higher than that in stage ?+ ?(P<0.05),the expression level of KLF4 in middle-low differentiation group was significantly lower than that in high differentiation group(P<0.05),the expression level of KLF4 protein in lymph node metastasis group was significantly lower than that in non-lymph node metastasis group(P<0.05);the expression level of Ki-67 protein in stage ?+? was significantly lower than that in stage ?+ ?(P<0.05),and the expression level of Ki-67 protein in middle-low differentiation group was significantly higher than that in highly differentiated group(P<0.05),the expression level of Ki-67 protein in lymph node metastasis group was significantly higher than that in non-lymph node metastasis group(P<0.05);however,the expression level of KLF4 protein and Ki-67 protein was not significantly correlated with age,sex and tumor size(P > 0.05).3.Statistical analysis showed that there was a negative correlation between KLF4 protein and Ki-67 protein expression in cholangiocarcinoma tissues(r=-0.376,P<0.01),which was statistically significant.Conclusions:1.Compared with normal tissues adjacent to malignant bile duct tumors,KLF4 protein expression rates in malignant bile duct tumors was lower and significantly lower than that in normal tissues adjacent to malignant bile duct tumors.The difference in positive rates was statistically significant.The expression of KLF4 protein is related to lymphatic metastasis,differentiation and stage of biliary malignant tumors.It suggests that it is involved in the occurrence,invasion and metastasis of cholangiocarcinoma.2.Compared with the normal tissues adjacent to cholangiocarcinoma,the expression rates of Ki-67 protein in malignant bile duct tissues increased,and was significantly higher than that in normal bile duct tissues adjacent to cancer.The positive rates of Ki-67 protein was significantly higher than that in normal bile duct tissues adjacent to cancer.The expression of Ki-67 protein was correlated with the degree of differentiation of cholangiocarcinoma cells,lymph node metastasis and tumor stage.These datas suggest that Ki-67 may be involved in the occurrence,invasion and metastasis of biliary malignant tumors.3.There was a negative correlation between the expression of KLF4 and Ki-67 protein in biliary malignant tumors.KLF4 and Ki-67 may be related to the proliferation,invasion and migration of malignant bile duct cancer cells.They may play an important role in the occurrence and development of cholangiocarcinoma cell carcinomas.Joint detection may be more effective in judging the biological behaviors of cholangiocarcinoma.
