| OBJECTIVE:The model of diabetic blood glucose fluctuation model was used as the research object to evaluate the effect of Shenqi compound on the islet function of rats,and to explore the therapeutic effect of traditional Chinese medicine on diabetes.METHODS:70 male Sprague-Dawley rats were used to construct a rat model of diabetic blood glucose fluctuation.The 50 rats with successful model were randomly divided into model group(M),Glucophage group(G),Chinese medicine high-dose group(SH),Chinese medicine middle-dose group(SM),and Chinese medicine low-dose group(SL),according to the level of blood glucose.In addition,10 healthy SD rats were used as a blank control group(C).During the modeling period,rats in each experimental group were given corresponding drugs for intervention.At the same time,the general condition of the rats was observed,and the blood glucose levels at five time points in one day were measured weekly,and the blood glucose fluctuation index(MBG,SDBG,LAGE)was calculated.Blood was collected from the abdominal aorta after 8 weeks of intervention.The levels of serum insulin and glucagon in rats were detected by ELISA.Pancreatic tissue was collected and HE staining was used to observe the pathological changes of islets.Islet α cells,β cells and their ratios were detected by immunohistochemistry.The apoptosis of islet cells was detected by TUNEL.Result:1.Compared with the model group,the mental state,hair color,drinking water,food intake,polyuria and body weight of the Chinese medicine each dose groups and Glucophage group were generally better,and the Chinese medicine each dose groups were superior to the Glucophage group.2.Compared with group C,MBG,SDBG and LAGE were significantly increased in group M,group G and the Chinese medicine each dose groups,and the difference was statistically significant(P<0.01),indicating that blood glucose fluctuation persisted.3.Comparison of serum insulin levels after 8 weeks of intervention.Compared with group C,the insulin levels in group M,group G and Chinese medicine each dose groups decreased,and P of group M<0.05,the difference was statistically significant.Compared with the group M,the group G and the Chinese medicine each dose groups were higher than the group M,and the P of SH group was<0.05,the difference was statistically significant.Comparison of serum glucagon levels,compared with group C,group M,group G and the Chinese medicine each dose groups were higher than group C,(P<0.01,the difference was statistically significant.Compared with group M,group G and the Chinese medicine each dose groups differences were not statistically significant.4.After 8 weeks of intervention,the number of a cells,the number of β cells,and the ratio of α/β in islets were compared.Compared with group C,there was no significant difference in the number of a cells between group M,group G and the Chinese medicine each dose groups(P>0.05).Compared with group M,there was no significant difference in the number of a cells between group G and the Chinese medicine each dose groups(P>0.05).Compared with group C,the number of β cells in group M,group G and the Chinese medicine each dose groups decreased,and P<0.01 in group M,the difference was statistically significant.Compared with group M,the number of P cells in group G and the Chinese medicine each dose groups was higher than that in group M,and SL group(P<0.05),the difference was statistically significant.Compared with group C,the ratio of α/β in group M,group G and the Chinese medicine each dose groups was higher than that in group C,among which group M was P<0.05,the difference was statistically significant.Compared with the M group,the α/β ratio of the G group and the Chinese medicine each dose groups was higher than that of the M group,and the difference was not statistically significant.5.Comparison of islet β cell apoptosis after 8 weeks of intervention.Compared with group C,the number of β apoptotic cells in group M,group G and the Chinese medicine each dose groups increased,and P<0.01 in group M,the difference was statistically significant.Compared with group M,group G and SH,SL group were less than group M,P<0.01,the difference was statistically significant.Compared with the M group,the SM group had P<0.05,and the difference was statistically significant.Compared with group C,the density of apoptosis-positive cells in group M increased,P<0.05,which was statistically significant.Compared with group C,there was no statistical significance in the group of the Chinese medicine each dose groups.Compared with group M,the density of apoptosis-positive cells in group G and the Chinese medicine each dose groups was lower than that in group M.Among them,the SM group was P<0.05,and the SL group was P<0.01,which was statistically significant.6.Islet pathological staining(HE,200X,Bar=50μm)observed that the islet tissue structure of group C was normal,and the islet cells of rats in group M,group G and the Chinese medicine each dose groups were damaged,but the drug intervention groups were lighter than group M.CONCLUSION:Shenqi compound can improve the general condition of diabetic blood glucose fluctuation model rats.Diabetic blood glucose fluctuation model rats have the characteristics of insulin deficiency and hyperglucagonemia at the serum hormone level.Islet cells have the characteristics of severe β cell damage,abnormal growth of a cells and increased apoptosis of islet cells.Shenqi compound can promote insulin secretion in rat model of diabetic blood glucose fluctuation,and has the effect of enhancing islet β cell anti-injury and inhibiting islet cell apoptosis to regulate α/β value in diabetic blood glucose fluctuation model. |
