Objective: Based on the theory of "Spleen Deficiency and Pathogenic Accumulation",to study the effects of Shenqi Compound prescription on ? cells function and blood glucose fluctuation in GK rats.Method: 30 GK rats divided into 3 groups after 4 weeks of adaptive feeding: Model group,Shenqi Compound Prescription group,and Sigliptin group,which were continuous intervened with drugs and fed with high fat feedhigh-fat diet for 8 weeks.Ten Wistar rats were set as blank control group.They were given ordinary feed as parallel control.With the construction of the model,drug intervention was given.The Shenqi Compound Prescription group was given Shenqi Compound Prescription Extract by gavage.The Sigliptin group were given Sigliptin suspension by gavage.The blank group and model group were given normal saline.The general state of rats was observed during drug intervention.blood glucose was measured once a week which inclouded five points of time in a day.Meanwhile,the mean daily blood glucose level(MBG),standard deviation(SDBG)and maximum fluctuation of blood glucose(LAGE,the difference between the highest and lowest intraday blood glucose)were used to measure the blood glucose fluctuation of rats in each group.Eight weeks later,MBG,SDBG and LABG were measured in each group.At the 8th week of the experiment,blood samples were collectedd and to measured insulin,lip,to assessed?-cell function.After blood collection,the thoracic and abdominal cavity were cut quickly.Pancreatic tissue was taken for histopathological examination(HE staining)and pancreatic beta cell apoptosis(Tunel fluorescence double staining).Result: After 8 weeks of intervention,GK rats in each group showed the characteristics of drinking more and eating less,wasting weight and so on,while Shenqi compound group was better than the model group in general.In terms of blood glucose fluctuation and blood lipid level,MBG,SDBG,LAGE,TC,TG in blank group was lower than that in GK group,while that in Shenqi compound prescription group and sieglitine group was lower than that in model group,and the level of serum TG in Shenqi compound group was lower than that in sieglitine group.In the aspect of insulin secretion,compared with the blank group,the average blood glucose level of GK rats in each group was lower than that in the model group(P < 0.01),while the average insulin level in Shenqi compound prescription group and sieglitine group was higher than that in the model group.In the evaluation of islet ? cell function,the QUICKI,HOMA-? and HOMA-IR index of GK rats in each group was significantly worse than that in blank group,while the islet ? function in Shenqi compound group and sieglitine group was slightly better than that in model group.In terms of pancreatic pathomorphology and islet ? apoptosis,islet inflammation and injury and islet ? cell apoptosis in Shenqi compound group were lower than those in model group and sieglitine group.Conclusion: Shenqi compound compound can improve the general state,islet ? cell function,and reduce pancreatic tissue inflammatory reaction and injury,inhibit islet ?cell apoptosis,and reduce the occurrence of Glycemic Excursions in type 2 diabetic rats.
