Expression And Effect Of PYCR1 On Colon Cancer | Posted on:2020-11-01 | Degree:Master | Type:Thesis | Country:China | Candidate:X Qin | Full Text:PDF | GTID:2404330590480070 | Subject:Internal Medicine | Abstract/Summary: | PDF Full Text Request | Objective: To observe the effect of pyrroline-5-carpoxylate reductase-1 on proliferation?migration and invasion of colorectal cells.Methods: The PYCR1 mRNA expression levels between colorectal tissues and normal tissues was elevated by RT-qPCR and immumohistochemical staining.In addition,mRNA and protein levels of PYCR1 were examined in four colorectal cell lines by RT-qPCR and Western blot,respectively.The recombinant plasmids were transfected into HCT116 and SW480 by Lipofectamine 2000 and the inhibition efficiency was determined via RT-qPCR and Western blot.Cell proliferation ?migration and invasion were detected by CCK8 assay?Colony formation and Transwell assay.Furthermore,the levels of protein E-cadherin ?N-cadherin?Vimentin??-catenin and c-Myc were measured by Western blot.Results:The expression of PYCR1 is upregulation in colorectal tissues and cell lines.Besides,PYCR1 mRNA(P<0.05)and protein(P<0.05)levels were significantly lower after shPYCR1 transfection Compared with the control shPYCR1-vector group,the shPYCR1 group was remarkablyinhibited in cell proliferation?migration and invasion abilities.At last,the expression levels of E-cadherin in shPYCR1 group was increased(P<0.05),but N-cadherin ?
