Objectives Based on Fine needle aspiration biopsy,the expression of Galectin-3,TPX2 and CCNG2 in benign and malignant thyroid nodules was detected,and the clinical value of Galectin-3,TPX2 and CCNG2 in the diagnosis of benign and malignant thyroid nodules was discussed.Methods The enrolled cases were from 108 patients with thyroid nodules treated surgically in Tangshan Workers’ Hospital and Tangshan People’s Hospital from November2017 to July 2018.Cytological smears were obtained from thyroid nodules after operation by simulated fine needle puncture in vitro.The expression of Galectin-3,TPX2 and CCNG2 in benign and malignant nodules were detected by immunocytochemistry.The results of immunocytochemistry were evaluated by semi-quantitative scoring method.Postoperative pathological findings were used as the gold standard for judging benign and malignant nodules.Evaluation of sensitivity,specificity,Yoden index and consistency of three molecular markers,Galectin-3,TPX2 and CCNG2,individually and jointly.Results 1 The expression rate of Galectin-3 in benign and malignant nodules was 7.89%vs 94.29%(P < 0.05),with significant difference.The expression rate of TPX2 in benign and malignant nodules was 10.53% vs 65.71%(P < 0.05),with significant difference.The expression rate of CCNG2 in benign and malignant nodules was 94.74% vs 32.86%(P <0.05),with significant difference.2 Multivariate analysis showed that Galectin-3(OR 0.005;95% CI 0.001-0.036)and CCNG2(OR 35.786;95% CI 3.776-339.138)were independent influencing factors for the diagnosis of benign and malignant thyroid nodules,while TPX2 could not be used as an independent influencing factor for the diagnosis of benign and malignant thyroid nodules(OR 3.065;95% CI 0.332-28.310;P=0.323).3 The sensitivity,specificity,Yoden index and Kappa value of Galectin-3 and CCNG2 were 94.29% vs67.14%,92.11% vs 94.74%,86.40% vs 61.88% and 0.859 vs 0.549 respectively.4.The Kappa value of Galectin-3 is significantly higher than that of CCNG2,so the sensitivity,specificity,Yoden index and Kappa value of Galectin-3 in series and parallel are 64.29%vs 97.14%,100% vs 86.84%,64.29% vs 83.98% and 0.559 vs 0.855,respectively.Conclusions 1 Galectin-3 and TPX2 were highly expressed in malignant tissues and CCNG2 was highly expressed in benign tissues.2 TPX2 has little value in the diagnosis of benign and malignant thyroid nodules.3 Galectin-3 was highly consistent with gold standard in the diagnosis of thyroid nodules,while CCNG2 was moderately consistent with gold standard in the diagnosis of thyroid nodules.4.The diagnostic value of Galectin-3 and CCNG2 in series was no better than that of Galectin-3 alone.The diagnostic value of 5Galectin-3 and CCNG2 parallel detection was significantly higher than that of Galectin-3alone.Figure 6;Table 7;Reference 47...
