The thesis topic is from the Research And Development Project Of Jiang xi Province?No.20171BBG70104?.Akebia trifoliata?Thunb.?Koidz is one of the traditional Chinese medicine in China.We studied the chemical constituent of Akebia trifoliata?Thunb.?Koidz and set up the quality standard.It is of great significance to clarify the chemical compositions and establish quality standard of Akebia trifoliata?Thunb.?Koidz.And our study provided important scientific basis for making full use of Akebia trifoliata?Thunb.?Koidz.In this dissertation,the following results were achieved.1.Study on the Chemical Constituents of Akebia trifoliata?Thunb.?KoidzThe columnl chromatography,Sephadex LH-20 and HPLC were used to isolate and purify the compounds of n-butanol Extract Part of the Akebia Trifoliata?Thunb.?Koidz.The chemical structures were identified on basis of physic-chemical properties and spectral data.Result:21 compounds were isolated identified as mutongsaponin B?1?,mutongsaponin C?2?,saponin PH?3?,Begoniifolide A?4?,2?,3?,23-trihydroxy-30-noroleana-12,19-dien-28-oic acid-O-?-D-xylopyranosyl-?1?3?-?-L-rhamnopyranosyl-?1?4?-?-D-glucopyranosyl-?1?6?-?-D-glucopyranosyl ester?5?,2?,3?,20?-trihydroxy-30-noroleana-12-en-28-oic acid-O-?-D-x ylopyranosyl-?1?3?-?-L-rhamnopyranosyl-?1?4?-?-D-glucopyranosyl-?1?6?-?-D-glucopyranosyl ester?6?,2?,3?,20-trihydroxy-30-noroleana-12-en-28-oic acid-O-?-L-rhamnopyranosyl-?1?4?-?-D-glucopyranosyl-?1?6?-?-D-glucopyranosylester?7?,Akemisaponins D?8?,Akemisaponins E?9?,asiaticoside?10?,saponin PJ1?11?,sch effoleoside A?12?,aketrilignoside B?13?,akemisaponins A?14?,aketrilignoside A?15?,symplocosneolignan A?16?,Kalopanax-saponins D?17?,Leonticin E?18?,ciwujia noside A1?19?,2?,3?,20?,23-trihydroxy-12-en-28-oic acid-?-L-rhamnopyranosyl-?1?4?-?-D-glucopyranosyl-?1?6?-?-D-glucopyrano-syl ester?20?,compound A?21?.Conclusion:Compounds 1<sup>4?16?18<sup>19 are isolated from this plant for the first time,Compounds20?21 are new compounds.2.Study on the Quality Standard of Akebia trifoliata?Thunb.?KoidzWe firstly established a TLC method for the identification of two Triterpenoid saponins chemical compositions in Akebia trifoliata?Thunb.?Koidz,namely,Saponin PH and saponin PJ1,using chloroform-methanol-formic acid?20?10?1?as the mobile,10%sulfuric acid ethanol solution was used as colorimetric agent.Results:The Saponin PH and saponin PJ1 in Akebia trifoliata?Thunb.?Koidz could be identified by TLC.Based on method validation,this method with good repeatability can be used for the TLC of Akebia trifoliata?Thunb.?Koidz.We established an HPLC method for the simultaneous determination of four Triterpenoid saponins chemical compositions in Akebia trifoliata?Thunb.?Koidz,namely,Saponin PH,Akemisaponins E,saponin PJ1 and Scheffoleoside A.The ELITE C18?250 mm×4.6 mm,5?m?was used with the mobile phase of acetonitrile-water?21?79?,at the flow rate of 1.0 mL·min-1,the detection wavelength was 200 nm,and the column temperature was at 30?.Results:The Saponin PH and saponin PJ1 in Akebia trifoliata?Thunb.?Koidz could be identified by TLC.The linear range of Saponin PH,Akemisaponins E,saponin PJ1and Scheffoleoside A were 0.20115.0275?g?r=0.9999??0.07551.8875?g?r=0.9999??0.22435.6075?g?r=1.0000?and 0.04141.0350?g?r=0.9999?,respe ctively.The average recoveries?n=3?were 99.80%?RSD=0.46%??98.25%?RS D=1.15%??99.5%?RSD=0.4%?and 98.88%?RSD<1.01%?,respectively.the precision and stability are good.The content of saponin PH,Akemisaponins E,saponin PJ1 and Scheffoleoside A in five batches of samples were 2.0943.928?0.9561.418?2.9584.472 and 0.5260.797 mg·g-1.Conclusion:This method is specific,accurate and reproducible,and can be used to control the quality of Akebia trifoliata?Thunb.?Koidz.
