Purpose:After several modifications,Atractylodes macrocephala Koidz. species for the Pharmacopoeia,Pharmacopoeia edition in 2005 has been included Characters, Identification,Check,Process and so on,but it is still a lack of Determination which is criteria for crude drugs and decoction pieces. This paper made Atractylodes macrocephala Koidz. as its study object,separated chemical components,and got the main ones or index ones,and established a determination method to control quality of Atractylodes macrocephala Koidz..At the same time,improving or perfecting some contents of quality standards of Chinese Pharmacopoeia in order to better control the quality of Atractylodes crude drugs and decoction pieces.Material and method : Use the method of solvent extraction , Silica gel column chromatography,Sephadex LH-20 column and recrystalizition;according to the method of the physical and chemical characteristics and spectral;by HPLC method, establish the method of the determination,the ash,water and extract of Atractylodes crude drug,decoction pieces and processing;by HPLC method;establish the fingerprints of Atractylodes decoction pieces and processing ones,and compare the two components.ResuLts:1. In this thesis,six compounds are separated from Atractylodes macrocephala Koidz.,which were 1)atractylenolideⅠ,2)atractylenolideⅡ,3)atractylenolideⅢ,4)taracetate,5)β-sitosterol, 6)stigmasterol.2. Take atractylenolideⅠ,Ⅱ,Ⅲas indicators , it is establisheded the determination method of Atractylodes crude drugs,decoction pieces and processing ones,and made the methodology study.The method is simple and accurate. Ten regions of Atractylodes crude drugs,twenty regions of decoction pieces and processing were determined by this method.3. Refering to Atractylodes crude drugs of Pharmacopoeia edition in 2005, Characters,Identification,Check and so on,established the above methods and limits for Atractylodes decoction pieces and processing;add Water, extraction methods of Atractylodes crude drugs,decoction pieces and processing;it is accurately revised and improved the quality of Atractylodes crude drugs,while updated decoction pieces and processing quality standard.4. AtractylenolideⅡis the main active components as Atractylodes indicators. To establish the preparation method and quality standard of atractylenolideⅡ,by solvents and chromatography,atractylenolideⅡwas isolated and prepared reference substances from Atractylodes.The structure was made by UV , IR , MS , 1H-NMR and13C-NMR.The purification and determination were made by TLC and HPLC.5. The range of similarity degree of HPLC fingerprint of Atractylodes decoction pieces and processing ones was between 0.90～1.00.Seventeen mutual peaks were selected in chromatography,and the most of the detected peaks were separated effectively;by comparing Atractylodes decoction pieces and processing ones, chemical ingredients are almost same.ConcLusion:1. Isolated compounds from Atractylodes macrocephala Koidz.and identified their stuctures.2. Made quality standards of Atractylodes decoction pieces and processing,including Characters,Identification,Check,Extract, Determination and so on.3. Improved quality standard of Atractylodes crude drugs, added Water, extraction methods of Atractylodes crude drugs ,and established the content determination.4. Established the fingerprint of Atractylodes decoction pieces and processing,by comparing,chemical ingredients are almost same.
