OBJECTIVE:1.To explore the effects of FTZ on energy metabolism by different animal models.2.From the perspective of energy metabolism as the entry point,the orthogonal design method was used to disassemble the FTZ to explore the effect of single-flavor and drug compatibility on energy metabolism in FTZ.Another idea for FTZ treatment of glycolipid metabolic diseases.METHORDS:Different animal models such as C57BL/6J normal mice,aged mice and chlorpromazine-induced cryopreserved mice were used to observe the effects of compound Qizhu Tiaozhi Capsule(FTZ)on energy metabolism.The C57BL/6J normal experimental group and the old experimental group were measured for anal temperature changes at 0h,1.5h,and 3.0h before and after administration on the 1st,4th,7th,and 14 th day after administration.On day 14 of dosing,the mice were placed in a comprehensive animal energy monitoring system to monitor the energy metabolism of the mice;On the 14 th day of administration,the chlorpromazine hypothermia experimental group was intraperitoneally injected with 0.08% chlorpromazine hydrochloride to measure the anal temperature and energy metabolism.Using the orthogonal design method of 13 factors and 3 levels,each medicine of FTZ was taken as a factor,three levels were set up,and the interaction groups of N and DZ、F and B were set up to obtain the orthogonal design.The medicinal materials were extracted according to the orthogonal design scheme,and body temperature and energy metabolism of mice after chlorpromazine modeling were measured according to the clinical equivalent dose of each disassembled group after 14 days,and the chlorpromazine model was established.After the mouse body temperature and energy metabolism.Taking the anal temperature,respiratory quotient,and oxygen consumption rate as indicators,the compound decoction was studied in the compound,and the main drugs that promote energy metabolism and their optimal drug compatibility were screened out.RESULTS:1.Short-term administration of FTZ can not change the body temperature,respiratory oxygen consumption rate and caloric value of aged mice and C57BL/6J normal mice.FTZ can promote the normal respiratory quotient of normal mice,but does not affect the respiratory quotient of aged mice.2..After intraperitoneal injection of chlorpromazine hydrochloride,the central body temperature of the mice decreased significantly,and the energy metabolism indexes such as respiratory oxygen consumption rate,respiratory quotient and caloric value decreased,and the duration was about 24 hours.3.After 2 weeks of administration of FTZ,the body temperature of chlorpromazine mice was relieved within 1 hour of modeling,and the oxygen consumption rate and respiratory quotient of chlorpromazine mice were improved.The best efficacy time is 12-16 h after modeling.4.8 drugs of FTZ had little effect on respiratory quotient of chlorpromazine mice,and had no significant effect.The anal temperature、 respiratory oxygen consumption rate and caloric value of chlorpromazine mice was greatly affected,among them,the N 、 H 、 N &DZ had significant effects on the oxygen consumption rate and caloric value of chlorpromazine mice(P<0.05).However,S、F、DZ、B、DS and DJ have slight effect of oxygen consumption rate and caloric value in hypothermia mice.N、DZ、and B all more affected by the dose,and the dose is larger,the effect on the oxygen consumption rate of chlorpromazine mice is more effecttive,while H is the opposite.The results of S at level 1 were similar to those at level 3,indicating that the effect of S on the oxygen consumption rate of chlorpromazine mice was different at different doses.The compatibility of N and DZ 、F and B is better than the effect of single-flavor medicine on its respiratory oxygen consumption rate.The best combination to promote energy metabolism is: N-24 g,DZ-10 g,F-10 g,B-24 g,S-0g,DJ-20 g,H-0g,DS-20g。 CONCLUTION:1.Chloramphenamine hydrochloride-induced mice have the characteristics of decreased body temperature and significant decrease in energy metabolism.2.Short-term administration of FTZ does not change the energy status of C57BL/6J normal mice and aged mice,but it can alleviate the decrease of body temperature caused by chlorpromazine hydrochloride and promote its energy metabolism.3.FTZ mainly promotes the energy metabolism of mice by increasing the respiratory oxygen consumption rate and caloric value of mice.Among them,N has the most significant effect on energy metabolism,followed by H and S,then followed by F、B and DZ,DS and DJ affect little.Among them,H mainly play a role in inhibition,and the N,DZ,and B mainly play a promoting role.N and DZ、F and B have a compatibility.
