Objective:Based on the regulation mechanism of miR-21/miR-122 to investigate the effect of Yiqi Huayu Jiedu Recipe on human hepatocarcinoma QGY7702/Sora resistant cell line and the possible mechanism of Yiqi Huayu Jiedu Recipe on the resistance of sorafenib to hepatocarcinoma,which provided scientific basis for clinical application of Yiqi Huayu Jiedu Recipe on the resistance of sorafenib to hepatocarcinomaMethods:QGY7702 cells cultured in vitro.QGY7702/Sora cell line resistant to sorafenib was induced by high dose shock and low dose maintenance.There were 5 groups in the experiment:blank group,model group,Yiqi Huayu Jiedu Recipe group,sorafenib group,Yiqi Huayu Jiedu Recipe combined with sorafenib group.CCK-8 method was used to detect the median inhibition concentration(IC50)and resistance index(RI)of sorafenib.The experimental group was intervened with IC50 concentration,the blank group and the model group were cultured with complete medium,and the cells were collected 48 hours after intervention.The expression levels of miR-21 and miR-122 were detected by RT-qPCR.Western blot was used to detect the expression of HIF-1,VEGF,E-cad and bcl-2Results:1.QGY7702/Sora resistant cell line was successfully cultured Sorafenib IC50 of parental cells and resistant cells respectively were(7.99±0.52)μmol/L and(19.65±1.22)μmol/L.RI was(2.49±0.28)2.Compared with the parent cells,the expression of miR-21 was overexpression and miR-122 was low expression(P<0.01).After drug intervention,the expression of miR-21 decreased significantly in all groups except sorafenib group(P<0.01),especially in Yiqi Huayu Jiedu Recipe group(P<0.01);the expression of miR-122 increased in each group(P<0.01),and the most significant effect was in Yiqi Huayu Jiedu Recipe group(P<0.01)3.The expression of HIF-1α,VEGF and Bcl-2 in the resistant cells was higher than that in the parental cells(P<0.05),while the expression of E-cad was lower(P<0.05).After drug intervention,the content of HIF-1αin each group decreased significantly(P<0.01),and the most significant inhibitory effect was in Yiqi Huayu Jiedu Recipe group(P<0.01);the expression of VEGF was significantly decreased in each group(P<0.01),and the most obvious inhibition was in the group of Yiqi Huayu Jiedu Recipe combined with sorafenib(P<0.05);the expression of E-cad in each group increased significantly(P<0.01),especially in the group of Yiqi Huayu Jiedu Recipe combined with sorafenib(P<0.01);The expression of Bcl-2 decreased in each group(P<0.05),and the most significant inhibitory effect was in Yiqi Huayu Jiedu Recipe group(P<0.01).Conclusion:Yiqi Huayu Jiedu Recipe can regulate the expression of miR-21/miR-122 by improving tumor hypoxia microenvironment,reduce the expression of VEGF,inhibit the angiogenesis,promote the expression of E-cad,inhibit the EMT process,inhibit the expression of Bcl-2,promote the apoptosis and other related effects to antagonize the resistance of human hepatoma QGY7702/Sora cells to sorafenib.
