| Objective: To investigate the difference of CD4+T lymphocyte in the pathogenesis of multiple sclerosis(MS)and neuromyelitis optica spectrum disorders(NMOSD)in neurological autoimmune demyelinating disease.Methods: 17 patients with recurrent MS,19 NMOSD patients and 10 normal controls were collected in this study.Peripheral blood was collected and PBMC and supernatant were separated.Flow cytometry was used to observe the changes of CD4 + T cells,including Th1,Th2,Th17,Treg cells,CD4 + i NKT cells and their intracellular cytokines IFN-γ,IL-4,IL-17,IL-10.The changes of cytokines IFN-γ,TNF-α,IL-15,IL-6,IL-10,IL-21,IL-23p19,IL-17 A,IL-1β,GM-CSF,G-CSF and TGF-α in supernatant were also detected.To screen trends of CD4+T cells and cytokines closely related to the pathogenesis and treatment of MS and NMOSD.The values of the three groups were compared.Results: 1.The secretion of IL-4 and IL-10 in MS and NMOSD group was lower than that in normal control group.2.IL-17 and INF-γ secretion in MS group and NMOSD group were more than that in normal control group.Among them,IFN-r secretion was more in MS group and IL-17 secretion was more in NMOSD group.The number of CD4 + CD25 + FOXP3 + Treg cells in the two groups was less than that in the normal group.3.The cytokines IL-1β,IL-6,TNF-α,IL-15,TGF-α and IL-23p19 in the supernatant of MS and NMOSD groups were higher than those in the control group.4.There were no significant differences in IL-10,IL-17 A,G-CSF,GM-CSF,IFN-γ and IL-21 among the three groups.Conclusion: 1.MS and NMOSD in Th1 and Th17 cells play a pathogenic role,but NMOSD tends to Th17 cell-mediated immune response compared with MS,and MS tends to Th1-mediated immune response compared with NMOSD.Both Treg and Th2 cells play an anti-inflammatory role.2.IL-1β,IL-6,TNF-α,IL-15,TGF-α,IL-23p19 play a positive role in the pathogenesis of MS and NMOSD. |
