Cyathulae Radix,the dried root of Cyathula officinalis Kuan,has long been used for eliminating congestion,stimulating the menstrual flow,easing movement of joint,promoting diuresis for relieving strangurtia,etc.Processed methods of Cyathulae Radix include cutting and stir-frying with wine,and their processed production is Cyathulae Radix slice?called Chuanniuxipian,CNXP in Chinese?and wine Cyathulae Radix slice?called Jiuchuaniuxi,JCNX in Chinese?,respectively.However,based on2015 Edition Chinese Pharmacopoeia?ChP?,the standards of JCNX and its processed method remain unclear.Traditionally,the processed production is identified by morphological characteristics which are difficult to ensure the quality.What's more,the differences between CNXP and JCNX are large.According to 2015 Edition ChP,the quality standard of Cyathulae Radix could not differentiate Cyathula capitata Moq.,Cyathula officinalis*Cyathula capitata and their adulterant.Thereby,a standardized,objective and modern processed technology for Cyathulae Radix and a systematical quality control that can identify adulteration are timely.Our research focuses on three aspects.Study on the processed technology of CNXP.In this section,we used eight HPLC characteristic peaks?including cyasterone?,extract,softening degree score and the ratio of forming of CNXP as markers to evaluate the influence of time on wash and soften process.we used ratio of forming of CNXP as markers to evaluate the influence of slice thickness on section process.we used eight HPLC characteristic peaks?including cyasterone?,extract,moisture of CNXP as markers to evaluate the influence of temperature and time on dry process.Through the evaluation results combined with the actual situation of modern processing,the optimal processed technology of CNXP is as followed:taking Cyathulae Radix,eliminating impurities,removing heads,and dividing them according to the maximum diameter of the middle part of Cyathulae Radix which are 1.8cm,washing and piling up by water,softening for 524h,cutting to 2±0.25mm,drying at 65±5°C for 1.52h,remove impurities.Study on the processed technology of JCNX.This section take alcohol,total sugar and?-phenyl ethanol combined with actual production costs as markers to optimize type of auxiliary rice wine for processed,and the best choice is traditional dry yellow wine or semi-dry yellow wine made from rice.Eight HPLC characteristic peaks?including 5-hydroxymethylfurfural,cyasterone?,extract,moisture and character are used as markers to investigate the relationship among the amount of alcohol added,the time of moisten,the proportion of the maximum amount of fried drugs,the temperature and time of stir-fry,and the speed of rotary drum with the characteristic of inside and outside of JCNX,end-point drug temperature.Based on the uniform design of information entropy theory,the regression analysis of the proportion of the maximum amount of fried drugs,the stir-fry temperature and the fired time is performed.Combined with the actual situation of modern processed,the optimal processed method of JCNX is as followed:taking CNXP,adding 10%yellow wine according to the proportion of CNXP and moistening for 23h,weighing the proportion of the maximum amount of fried drugs of 40±5%,setting the rotation speed to 15±2r·min-1,using drum-type speculum machine with a constant temperature of 200±10°C,and fired them for 15±2min.When the temperature of CNXP reaches135±5°C,taken them out,make cool,remove impurities.Study on the quality standards of Cyathulae Radix,CNXP and JCNX.This section mainly studies the quality standard of Cyathulae Radix,CNXP and JCNX.Combed character,revised the TLC method,add foam identification and?-phenyl ethanol identification,revised the extract method.Meanwhile.A new HPLC specific chromatogram combined with content determination?5-hydroxymethylfurfural,cyasterone?method are added,and a new range of limits is developed.At the same time,study on moisture and total ash is also carried out.In addition,the method for determination of ferulic acid is also studied,but it is also detected in the counterfeit productions of Cyathulae Radix and the sample preparation method is complicated.So,it is not included in quality standard.According to our research,the quality standards of Cyathulae Radix,CNXP and JCNX are drafted.In summary,this project has studied the CNXP and JCNX processed techniques and the quality standards of Cyathulae Radix,CNXP and JCNX.Our research also provides a scientific basis for standardizing the processed technology of Cyathulae Radix and the identification of‘authenticity and raw and fried'.We also afford a reference for the in-depth study of the mechanism of JCNX.
