Ligusticum (Chuanxiong Hort). Having been crashed for dry rhizome of Chuanxiong (Ligusticum Chuanxiong Hort.) in common temperature, it got total sugar of Chuan xiong (Ligusticum Chuanxiong Hort.) by extracting and undoing liposolubility materials with cold water, diffusing with heat water , and condensing with water-bath. In term with the property that polysaccharide never dissolve in 95% alcoholic solution, it precipitate crude polycose. After anhydration by absolute alcohol, absolute ether, and actone, andvacuum dehydration, the crude polysaccharide powder of Chuanxiong (Ligusticum Chuanxiong Hort.) has been made. Through weighing calculation, the yield of crude polysaccharide was 6.7%. After removed protein by the combined deprotionization methods of enzyme and sevage and fractionated by ultra filtration and gel chromatography, the polysaccharide (LPC-Ⅳ) was obtained. Homogeneity and molecular weight of LCP-Ⅳwere determined by Gel filtration on Sepharose CL-6B, specific optical rotation measurement and High-performance liquid chromatography (HPLC) analysis. The sugar composition was determined by methanolysis and gas chromatography (GC). The results revealed that LPC-Ⅳis homogeneous on both molecular weight and polarity. The molar weight is estimated to be 3.2kd by means of HPLC.Through IR, GC, partial hydrolysis with acid, periodate oxidation, smith degradation, methylation and GC-MS analysis, the result indicate that LPC-Ⅳhas a kind of lowly branched structure. The main chain is mainly made up of major(1→6)Glc and some(1→4,6)Glucose and (1→3,6)Man; The side chain is composed of (1→6)Gal, (1→4) Gal and (1→3)Ara; The nonreduced end is composed of glucose. Residues are substituted at 4-O of (1→4,6 ) Glc and Man(1→3,6), On average, there are about 2 branches among 10 residues.
