Objective:To investigate the effects of scalp acupuncture combined with riched environmental stimulation on the activities of AchE and ChAT in hippocampus of rats with Alzheimer's disease,and to explore the possible mechanism and theory of scalp acupuncture combined with riched environment in treating AD.It provides reliable methods and theoretical basis for its clinical application.Methods:Alzheimer's disease model was induced by directed injection of A? 1-42 into the CA1 region of hippocampus in rats.72 wistar rats were randomly divided into six groups:sham operation group,model group,scalp plexus acupuncture group,enriched environment group,scalp acupuncture combined with riched environment group(combined group),western medicine group(12 rats each).The rats in the sham operation group were injected with the same volume of distilled water in the bilateral hippocampus,and the other groups were injected with A?1-42 in the bilateral hippocampus.The sham operation group and the model group were given the same grip and feeding after the intervention of the model,and no other treatment was given.The scalp point cluster acupuncture group was treated with scalp point cluster acupuncture,the rich environment group was given rich environment treatment,the western medicine group was given Anlisin gavage therapy,and the combined group was combined with rich environmental stimulation therapy on the basis of scalp point cluster acupuncture group.Results:1.Water maze: Escape latency:after four days of training,the escape latency of rats on the fifth day was as follows:sham operation group<Anlisin group<scalp acupuncture combined with rich environment group<scalp plexus acupuncture group<rich environment group<model group.Second quadrant percentage and number of crossing platforms:after 5 days of positioning training,The results of the second quadrant percentage and the times of crossing the platform on the 5th day were as follows:model group<rich environment group<scalp acupuncture combined with rich environment group<Anlisin group<sham operation group2.Histopathological changes of hippocampus:Sham operation group:the hippocampus area nerve cell disperses closely,the neuron shape basically does not change is relatively more regular.Model group:nerve cells scattered in the hippocampus lack of close and dense trend,relatively disordered,neurons have shown irregular and complete morphological trend,the number of glial cells has increased to a large extent,and the number of glial cells has increased greatly,and the number of glial cells has increased greatly At the same time,a large number of neuronal apoptosis in the picture is obvious.Scalp point plexus acupuncture group,rich environment group,scalp point cluster acupuncture combined with rich environment group:three groups of rats had a great optimization in the shape of the hippocampus:the distribution of nerve cells in the hippocampus was relatively close,and the loose morphology was obviously improved.Compared with the model group,the number of inflammatory cells,nucleated cells and glial cells decreased to a certain extent.3.The active expression of ChAT and AchE:The results of ChAT activity in hippocampus were as follows activity of model group<activity of Anlisin group<activity of rich environment group<activity of scalp point from acupuncture group<activity of cluster acupuncture of scalp point<activity of sham operation group.AchE activity in hippocampus:model group activity>rich environment group activity>scalp acupuncture plus rich environment group activity>Anlisin group activity>sham operation group activity.Conclusion:1.Cluster Needling of Scalp Point Therapy combined with enriched environmental can improve the learning and memory ability of AD rats,which is better than that of simple scalp acupoint cluster needling group or enriched environment group.2.Cluster Needling of Scalp Point Therapy combined with enriched environmental can improve the pathological morphology of hippocampus in AD rats,which is superior to simple Scalp-point cluster needling group or rich environment group.3.The increase of ChAT and the down-regulation of AchE activity in hippocampus and cortex of AD rats induced by scalp acupuncture combined with enriched environmental stimulation may be one of the related mechanisms in the treatment of AD,and the combination of scalp acupuncture and riched environmental stimulation has better therapeutic effect.It was obviously better than simple clump prickles or simple rich environmental stimulation.
