OBJECTIVEThe osteogenesis and antimicrobial activities of beta-tricalcium phosphate(beta-TCP)containing 10% silver nanoparticles were studied by the animal experiments.METHODSThe biocompatibility of ?-TCP containing 10% silver nanoparticles(?-TCP-Ag)was evaluated by the cytotoxicity test in vitro,the histological study of vital organs and the detection of silver ion in the tissue.The ?-TCP-Ag(?-TCP-Ag)and pure ?-TCP(?-TCP group)were implanted into the femoral condyle defects with osteomyelitis in the New Zealand white rabbits.After the operation the rabbits in the latter group was treated with penicillin(?-TCP(+))or without antibiotics(?-TCP(-)).The healing process of the infected bone defects was observed.RESULTSThe extract solution of ?-TCP-Ag had no effect on the proliferation of rBMSCs in vitro cytotoxicity experiments.Twelve weeks after the implantation,the contents of silver ions in heart,liver,spleen,lung,kidney and blood in the ?-TCP-Ag group and ?-TCP group were less than 0.1 ppm.There was no pathological changes in the HE staining sections of heart,liver,spleen,lung and kidney in the two groups.Twelve weeks after the implantation of ?-TCP-Ag into the femoral condyle defect with osteomyelitis in the rabbits,the micro-CT showed that the ratio of bone tissue mass to total bone volume(BV/TV)in the defect area was higher in the ?-TCP-Ag group than that in the ?-TCP group.The VG staining showed that new bone tissue grew into the pores of the ?-TCP-Ag group.But no obvious bone tissue was found in the pores of the ?-TCP(-)group.CONCLUSIONGS?-TCP containing 10% silver nanoparticles had good biocompatibility.After the implantation into the rabbit femoral condyle defect with osteomyelitis,it could effectively control the infection and promote the healing of bone defect,and had no obvious toxic and side effects on the vital organs.It is hopeful to provide a new material for the treatment of bone defects with infection or potential infection.
