Objective:To investigate the expression of Eps8,E-cadherin and Vimentin protein in different cervical tissues,and explore the correlation between the expression of Eps8 and the progression as well as epithelial–mesenchymal transition(EMT)of cervical cancer.Inhibit the expression of Eps8 in cervical cancer cells,and investigate the effects of Eps8 knockdown on the expression of EMT–related markers and the invasive and migration capacity of cervical cancer cells,to explore the role Eps8 plays in EMT process of cervical cancer.Methods:The expression of Eps8,E-cadherin and Vimentin protein in normal cervical,cervical intraepithelial neoplasia(CIN)Ⅰ,CINⅡ-Ⅲ and cervical squamous cell carcinoma tissues were detected by immunohistochemistry.The expression levels of these three proteins in different cervical tissues were compared,we further analyzed the correlation between the expression of Eps8,E-cadherin and Vimentin protein in cervical squamous cell carcinoma tissues.The correlation between the expression levels of these three proteins and the clinicopathological characteristics of cervial cancer were also analyzed.Liposome transfection technique was applied to inhibit the expression of Eps8 in cervical cancer cells,qRT-PCR,Western blot were used to detect the expression changes of E-cadherin,Vimentin and Snail caused by inhibition of Eps8 expression.The transwell chambers were used to carry out invasion and migration assays to analyse the effect of Eps8 knockdown on the capacity of invasive and migration in cervical cancer cells.Results:1.The expression of Eps8 and Vimentin in cervical squamous cell carcinoma tissues were higher than normal cervical tissues,CIN tissues;The expression of E-cadherin in normal cervical tissues,CIN tissues were higher than cervical squamous cell carcinoma tissues(P<0.05).2.In cervical squamous cell carcinoma tissues,there was a significant negative correlation between expression of E-cadherin and Eps8,as well as between expression of E-cadherin and Vimentin(P<0.05).The expression of Eps8 was positively related to that of Vimentin(P<0.001).3.The expression of Eps8 in squamous cell carcinoma tissues was significantly related to tumor differentiation degree,depth of infiltration and lymph node metastasis.The expression of E-cadherin and Vimentin were related to depth of infiltration and lymph node metastasis(P<0.05).4.qRT-PCR and Western blot analysis demonstrated that,the expression of E-cadherin was significantly increased,while the expression of the Vimentin,as well as Snail,was significantly decreased,which were caused by downregulation of Eps8 in cervical cancer cells(P<0.05).5.The transwell invasion and migration assays revealed that Eps8 knockdown statistically inhibited the invasive and migration capacity of cervical cancer cells(P<0.05).Conclusions:1.Eps8 may participate in the development,invasion and metastasis of cervical cancer.2.Eps8 may play an important role in EMT process of cervical cancer by regulating the expression of E-cadherin and Vimentin.3.Inhibition of Eps8 impairs the process of EMT in cervical cancer and notably inhibited the invasive and migration capacity of cervical cancer cells,Eps8 may promote cervical cancer metastasis by orchestrating EMT.
