Background: Cerebral infarction is one of the main causes of long-term dysfunction,which reduces the patientundefineds independence and quality of life.Previous studies have demonstrated the role of nonspecific inflammation in ischemia/reperfusion injury.Some studies have shown that interleukin is associated with atherosclerosis and plays an important role in cerebrovascular diseases.Il-101082A/G polymorphism affects the risk of high-risk factors of cerebral infarction such as diabetes,hypertension,smoking and so on.In view of the key role of single nucleotide polymorphism sites-1082 and-819 in the promoter region of IL-10 gene,the authors speculate that they are related to the incidence of cerebral infarction.This study focused on the relationship between promoter polymorphism of IL-10 gene and cerebral infarction.Amplification mutation system-polymerase chain reaction(ARMSPCR)was used to detect the distribution frequency of-1082 and-819 in patients with cerebral infarction.Objective: To investigate the association between interleukin-10 promoter gene polymorphism and cerebral infarction.Methods: Research methods were developed in strict accordance with the clinical epidemiological case-control study standard protocol.According to the calculation formula of sample size,the required sample size is calculated.From July 2016 to July2017,269 patients with acute cerebral infarction admitted to the Department of Neurology,second people's Hospital of Nanyang,were selected as observation group.A total of 538 non-cerebral infarction patients with high risk factors for cerebral infarction who were admitted to the stroke screening clinic at the same time were selected as the control group.Patients in the observation group and the control group were matched according to sex and age according to the sample size 1:2.At the same time,the high risk factors of cerebral infarction(hypertension,diabetes mellitus,coronary heart disease,history of atrial fibrillation,etc.)were collected and genotyped by polymerase chain reaction(PCR).Results: There was a significant difference in the genotype distribution and allele frequency of IL-10-1082 A/G allele between the observation group and the control group(? 2=6.643,P=0.01).The IL-10-1082 A allele frequency in the observation group was significantly higher than that in the control group(P=0.015).The IL-10-1082 AA genotype has a 2-fold higher risk of cerebral infarction than the AG genotype.IL-10-1082 AA genotype and increased fibrinogen,high homocysteine is an risk factor for cerebral infarction(OR = 2.073,95% CI: 1.278 ~ 3.364).There was no significant difference in IL-10-819 C/T genotype distribution and allele frequency between the observation group and the control group(P>0.05).Conclusion: IL-10-1082 A/G gene polymorphism is associated with cerebral infarction,and the increase in A allele frequency is closely related to the occurrence of cerebral infarction.
