| BackgroundPGRN is a secreted multifunctional glycoprotein growth factor,whose main function is to participate in the process of inflammation and angiogenesis.It can regulate the cell proliferation,migration,survival and take part in tumorigenesis,wound healing and nurodegradation-related diseases and so on.Overexpression of PGRN in living tissue can affect angiogenesis.Meanwhile,it is reported that PGRN plays an important role in early embryonic development and tissue remodeling.Objective:To explore the effect of PGRN on the blood vessels in early embryo and late placenta.Methodscollect placenta tissue of pre-eclampsia(10 cases)and health(12 cases)by caesarean section in Chongqing Medical University affiliated to the first hospital during January to April in 2017;detect the expression of PGRN in pre-eclampsia and control group by PCR and western blot;detect the blood vessels separately in 6.5-day embryo and 17.5-day placenta in PGRN-/-and WTmicebyhematoxylin-eosinandimmunohistochemicaland immunofluorescence staining.ResultsPGRN was expressed in pre-eclampsia and control group,and expression in pre-eclapsia.The number of embryo decreased and the development of the embryo was slower in PGRN-/-mice at E6.5(embryonic day of 6.5).Meanwhile,the expression of vascular marker SMA(smooth muscle actin)in PGRN-/-mice was lower than WT mice.It indicates that PGRN may affect embryonic vascular in early pregnancy.The diameter and the muscle thickness of uterine artery in PGRN-/-mice deceased compared with WT mice.It reminds us PGRN may play a role in uterine artery.The expression of blood vessel markers,like Tie2,SMA,COX2 in PGRN-/-placental labyrinth reduced compared with WT mice at E17.5.It suggests that PGRN may influence the development of placental vascular.Conclusionthe decline PGRN will affect the vascular development in embryo and placenta which behaves as a reduction in angiogenesis. |
