Objectives To count and analyze the relationship between CXCL12,CXCR4,VEGF,PLGF and the incidence of preeclampsia and its combined FGR by detecting the levels of these four factors in maternal serum and placental tissue,so as to explore the role of these four factors in the pathogenesis of preeclampsia and its combined with fetal growth restriction.Methods Pregnant women who were hospitalized in the Maternal and Child Health care Hospital of Anhui Province from May 2018 to December 2019 were selected as the object of study.62 pregnant women diagnosed as pre-eclampsia were pre-eclampsia group(PE group),among them,34 pregnant women who did not have fetal growth restriction(PE-FGR group)and 28 pregnant women with fetal growth restriction(PE+FGR group).51 normal pregnant women who were hospitalized at the same time were normal control group(N group).Collecting the general clinical data of the object of study,and collecting the prenatal maternal blood samples and placental tissue during delivery.The levels of CXCL12,CXCR4,VEGF and PLGF in the maternal serum were detected by ELISA,the levels of CXCL12,CXCR4,VEGF and PLGF in the placental tissue were detected by immunohistochemistry.Using t test,one-way analysis of variance,and?~2test to analyze the difference of the levels of CXCL12,CXCR4,VEGF and PLGF in maternal serum and placental tissue of each group.Results 1.The statistical analysis of the general clinical data:there were no statistically significant differences in the age,height,weight before pregnancy and number of pregnancy between PE group,PE-FGR group,PE+FGR group compared with N group(all P>0.05).The gestational weeks from high to low were N group(38.9±0.78w),PE-FGR group(36.9±2.59w)and PE+FGR group(32.4±2.66w),the differences were statistically significant(all P<0.05);The birth weight of newborn from high to low were N group(3.408±0.42kg),PE-FGR group(2.970±0.76kg)and PE+FGR group(1.457±0.41kg),the differences were statistically significant(all P<0.05).2.The results of Enzyme-linked immunosorbent assay showed that:the levels of CXCL12,CXCR4,VEGF and PLGF in the maternal serum of PE group were lower than those of N group,and the differences were statistically significant(t=10.18,17.86,12.92 and 7.49,all P<0.05).The levels of CXCL12,CXCR4,VEGF and PLGF in the maternal serum of the N group,PE-FGR group and PE+FGR group showed statistically significant differences(F=61.42,187.53,89.99 and 31.40,all P<0.05).The the levels of CXCL12 from high to low were N group(2509.45±160.20pg/ml),PE-FGR group(2241.97±195.03pg/ml)and PE+FGR group(2107.33±123.37pg/ml).The the levels of CXCR4 from high to low were N group(2225.01±105.77pg/ml),PE-FGR group(1935.92±90.16pg/ml)and PE+FGR group(1844.04±63.03pg/ml).The the levels of VEGF from high to low were N group(487.22±32.06pg/ml),PE-FGR group(425.51±26.88pg/ml)and PE+FGR group(408.30±19.33pg/ml).The the levels of PLGF from high to low were N group(259.82±16.47pg/ml),PE-FGR group(236.34±20.83pg/ml)and PE+FGR group(225.27±23.71pg/ml),and the levels of these four factors in the PE+FGR group were significantly reduced,the differences were statistically significant.3.The results of immunohistochemistry showed that:CXCL12,CXCR4,VEGF and PLGF are expressed in the cell membrane and cytoplasm of trophoblast cells,but VEGF and PLGF are also expressed in the vascular endothelial cells.In the placental tissue of the normal control group,CXCL12,CXCR4,VEGF and PLGF showed significant positive staining,the staining of CXCL12,CXCR4,VEGF and PLGF in PE group,PE-FGR group and PE+FGR group was relatively weakened.The levels of CXCL12,CXCR4,VEGF and PLGF in the placental tissue of PE group were lower than those of N group,and the differences were statistically significant(?~2=40.61,52.19,50.95 and 43.58,all P<0.05).The N group,PE-FGR group and PE+FGR group were compared in pairs,the levels of CXCL12,CXCR4,VEGF and PLGF in the N group,PE-FGR group and PE+FGR group were decreased successively,and the levels of these four factors in the PE+FGR group were significantly reduced,the differences between groups were statistically significant(all P<0.0167).Conclusions The levels of CXCL12,CXCR4,VEGF,and PLGF in maternal serum of patients with preeclampsia were reduced,and the levels of CXCL12,CXCR4,VEGF,and PLGF in placental tissue were also reduced,and the PE group combined with FGR was more significantly reduced;CXCL12,CXCR4,VEGF and PLGF may be important factors in the pathogenesis of preeclampsia and its combined FGR.
