Objectives To observe the protective effect of huoxue tongluoli fang on retinal nerve fiber layer of rats with acute retinal ischemia reperfusion(RIR)injury,as well as on aquaporin-4(AQP4),l-gluta-mate/l-aspartate transporter,GLAST,Also known as EAAT1)and the expression of glutamine synthetase(GS).Methods Experiments 1: 25 BN male rats were randomly selected to establish high intraocular pressure model.Optical coherence tomography(OCT)was used to observe the changes of retinal nerve fiber layer thickness with time in 5 BN rats,and to find out the regularity of the changes.According to the above experimental results,6 hours and 8 weeks were selected as two time observation points,and 20 BN rats were selected.Male rats were randomly divided into four groups: normal control group,model control group,traditional Chinese medicine control group and Ginkgo biloba leaf control group.The rats were administrated with the model.Then the changes of retinal nerve fiber layer thickness with time were observed by OCT to detect the effect of traditional Chinese medicine and Ginkgo biloba leaf.Experiment 2: 28 SD male rats were randomly divided into 7 groups.The first group was the normal control group,and the other six groups were the model control group.The model control group selected rats at 6 hours,12 hours,1 day,3 days,1 week and 2 weeks after the model establishment.Then the retinal tissues were extracted by anesthesia at 6 time points,and the most significant time points of AQP4,GLAST and GS expression were found by Western Blot experiment.2.According to the above results,6 hours after the establishment of the model were chosen as the time observation point;16 SD male rats were randomly divided into four groups: model control group,traditional Chinese medicine control group and ginkgo leaf control group.After 6 hours of administration of the model,rats were killed by anesthesia,retinal tissue was extracted,and Western Blot test was carried out to compare with the normal control group.The expression of AQP4,GLAST and GS in model control group,traditional Chinese medicine control group and ginkgo leaf control group.Results Experiments 1: 1At 6 hours,the thickness of nerve fiber layer increased,indicating that it was still in the edema stage,but decreased at 4,8 and 12 weeks,indicating that the nerve fiber layer became thinner due to high intraocular pressure in rats.After intervention with Chinese medicine and Ginkgo biloba leaves,it was found that the thickness of nerve fiber layer became thinner at 6 hours and lighter at 8 weeks.Experiments 2: 1 The most significant time points of expression of AQP4,GLAST and GS were 6 hours,while the other time points were not significant.2 At 6 hours,the expression of AQP4 in retina tissue of model control group was the lowest.The expression of AQP4 in traditional Chinese medicine group and ginkgo leaf control group was higher than that in model control group,but lower than that in normal control group.There was no significant difference between traditional Chinese medicine group and ginkgo leaf control group.The expression of GLAST and GS in traditional Chinese medicine group and ginkgo leaf control group was lower than that in model control group.But higher than the normal control group,there was no significant difference between the traditional Chinese medicine group and the ginkgo leaf control group.Conclusions 1 Huoxue tongluolishui decoction can reduce the degree of retinal edema and the thickness of retinal optic nerve fiber layer at the initial stage of retinal ischemia reperfusion injury;Long-term observation can reduce the thinning of optic nerve fiber layer and protect the optic nerve.Huoxue tongluo lishui can increase the expression level and activity of AQP4,GS and GLAST after retinal ischemia reperfusion injury,thus playing a role in protecting retinal tissue.2 Huoxue tongluo lishui can increase the expression levels and activity of AQP4,GS and GLAST after retinal ischemia reperfusion injury,thus protecting retinal tissue.Fig.5;Table 9;References 81.
