Objective: Analysis of the expression profile and role of circular RNA in Hepatocellular carcinoma?HCC?which provides a new basis for the diagnosis and treatment of HCC.Methods: 1.A total of 39 pairs of HCC tissues and adjacent non-tumor tissues?NCMT?were collected,and 5 pairs of homogenous tissue samples were selected.The circular RNA with the largest differential expression was hsacirc0000417 which was screened by circRNA chip technology,and its expression was verified by qPCR.2.Expanded sample testing to verify the expression of hsacirc0000417 in 34 HCC tissues and adjacent non-cancerous tissues,and at the same time verify the HCC cell lines 3.The virus knocking down the circular RNA was constructed and transfected into human hepatoma cells hepG2 and BEL7402,respectively.The function of the circular RNA was examined by plate cloning experiments,flow cytometry experiments,MTT,and apoptosis assays.Results: 1.Through the circRNA chip technology,304 of thecircular RNAs differentially expressed in HCC tissues were screened,of which 162 were up-regulated and 142 were down-regulated,of which hsacirc101099?circABasehsacirc0000417?was significantly up-regulated by about 12 times?FC=11.875?in HCC tissues.The results of qPCR showed that the expression of hsacirc0000417 in HCC was up-regulated by 7.60 times compared with NCMT.2.The qPCR verification results of expanding the sample showed that hsacirc0000417 was significantly higher in human hepatocarcinoma tissues than adjacent tissues,and also higher in hepatoma cells than in hepatocytes.The differences were statistically significant?P <0.05?.3.Compared with the control group,the human hepatoma cells hepG2 and BEL7402 transfected with the circular RNA virus were significantly decreased,the G2 phase arrest and the apoptotic rate were increased?P<0.05?.Conclusion: 1.Hsacirc0000417 is highly expressed in human HCC tissues and HCC cells,and it is expected to be a molecular marker for prognosis of HCC.2.Knocking down the expression of the circular RNA can inhibit the proliferation of human hepatoma cells,promote apoptosis,and interfere with the cell cycle,resulting in a significant increase in the proportion of cells in the mitotic phase?G2 phase?;it can be used as a potential target for targeted therapy...
