Objective: To observe the influence of Q-switched 1064 nm laser on chloasma guinea pig model before and after treatment of melanocyte(MC),providing animal experimental basis for combined treatment of chloasma.Methods: Twelve guinea pigs were randomly selected from 60 guinea pigs as group A(blank control group),and the remaining 48 guinea pigs were selected from group B(model preparation group).Samples were taken 45 days after model preparation to verify the success of model construction.The guinea pigs were randomly divided into four groups: B1 group(model control group),B2 group(laser treatment group),B3 group(soybean treatment group),and B4 group(soybean combined laser group).Treatment was performed on B2,B3 and B4 groups respectively.Skin Melan A immunohistochemistry was performed at different time points.Pathological image analysis system was used to record data.Before and after treatment,skin Superoxide Dismutase(SOD)activity and Malondialdehyde(MDA)concentration were determined.Results:(1)building,group B compared with group A,the SOD activity decreased(P<0.001),MDA content increased(P<0.001),the MC number density increased significantly(P<0.001),the MC positive goal number significantly higher(P<0.001),MC average grey value significantly reduced(P<0.001),the MC average optical density value was significantly increased(P<0.001),suggesting the guinea pig model of chloasma preparation of success.(2)there were differences in SOD activity and MDA content before and after treatment in B2,B3 and B4 groups(P<0.001),indicating that the three treatment methods were effective and had anti-free radical effect.F=9.473,P<0.001),indicating that there were significant differences between each group,and there were differences between two groups(P<0.001),and B4 group had the most significant effect of combined treatment.(3)since 5 weeks treatment immediately,B2,B3,B4 group compared with before treatment,the MC number density decreased significantly(P<0.01),the MC positive goal number decreased significantly(P<0.01),and a marked increase in the MC average grey value(P<0.01),the MC average optical density value decreased significantly(P<0.01),and 3 weeks after the end of treatment,with statistical significance(P<0.01).Compared with B2 and B3 groups,MC related data in B4 group showed the most significant changes(P<0.01),so B4 group had the most significant effect of combined treatment.Conclusion:(1)the use of "ultraviolet radiation" + "progesterone impact method" can be prepared chloasma guinea pig model;(2)soybean extract has anti-free radical effect;(3)nano-needle introduction of soybean extract on the chloasma guinea pig model has played a good transdermal absorption effect;(4)the Q1064 nm laser with large spot low energy mode has a certain therapeutic effect after several times of treatment has reached the threshold;(5)Q1064nm laser combined with nano-needle introduction of soybean extract treatment is more effective than single treatment.
