Objective: To evaluate the effect of human cord blood neural stem cells(HUCB-NSCs)on arterial transplantation in rats with cerebral hemorrhage by neuronal behavioral examination Longa score and microPET-CT,and to explore its possible therapeutic mechanism.Methods: The healthy adult Wistar rats were selected,and the model of rat cerebral hemorrhage was made by double blood injection/retraction method.The rats with successful modeling were randomly divided into experimental group and control group.Rats in experimental group were transplanted with human umbilical cord blood neural stem cells(HUCB-NSCs)via left ventricle.Rats in control group returned naturally without treatment.Neurological behavioral examination Longa score and microPET-CT scan were used to evaluate the recovery of nerve function,metabolic changes of brain injury site and volume changes of brain injury site in experimental group and control group respectively before transplantation and 3,7,14 and 21 days after transplantation.Results: 1.The longa score of arterial transplantation group and control group decreased gradually with time,differences were statistically significant(F artery = 774.889,P artery =0.000 < 0.05;F control = 184.657,P control = 0.000 < 0.05);the longa score of arterial transplantation group decreased more significantly than that of control group at the same time point,except before transplantation,at other time points,the longa score had statistical difference(P < 0.05).2.The hematoma and surrounding volume of rats in the two groups were enlarged on the 3rd day after transplantation.On the 7th,14 th and 21 st day after transplantation,the hematoma and surrounding volume of rats in the two groups decreased gradually with time,and there were significant differences at different time points(F artery =124.844,P = 0.001 < 0.05;F control = 135.685,P = 0.001 < 0.05).Compared with the control group at the same time point,the hematoma and surrounding volume of rats in the arterialtransplantation group decreased more significantly than those in the control group.Except for the pre-transplantation and the third day after transplantation,there were significant differences in hematoma and surrounding volume between the two groups at the remaining time points(7,14,21 days after transplantation)(P < 0.05).3.The level of glucose metabolism in microPET-CT scanned hematoma center of rats in arterial transplantation group decreased on the 3rd day after transplantation,and then increased gradually with time,the difference was statistically significant(F=11.648,P=0.001<0.05).The level of glucose metabolism in microPET-CT scanned hematoma center of rats in control group also decreased on the 3rd day after transplantation,and then also increased gradually with time,the difference was statistically significant(F = 11.661,P = 0.001 < 0.05).Compared with the control group at the same time point,except before transplantation and the third day after transplantation,the glucose metabolism level of the arterial transplantation group was higher than that of the control group at other time points,and the difference was statistically significant(P < 0.05).4.The level of glucose metabolism around hematoma in arterial transplantation group increased gradually with time,and the difference was statistically significant(F = 32.205,P = 0.001 < 0.05);The level of glucose metabolism around hematoma in control group increased gradually with time,and the difference was statistically significant(F = 11.116,P = 0.001 < 0.05).Compared with the control group at the same time point,except for pre-transplantation and the third day after transplantation,the glucose metabolism level of the arterial transplantation group was higher than that of the control group at other time points,and the difference was statistically significant(P < 0.05).Conclusions: The experimental results showed that HUCB-NSCs transplantation via arterial route could promote the recovery of nerve function,reduce the volume of hematoma and improve brain metabolism in rats with cerebral hemorrhage.
