Objective The aim of this study was to analyze the peripheral blood, liver and spleen lymph nodes, humoral immunity and cellular immune characteristics of patients with brucellosis in Hohhot,to understand the clinical characteristics of patients with Brucella infection, and to explore the immune response of Brucella after infection. The impact of the disease provides an experimental basis for the diagnosis and treatment of the disease in the future, reducing the rate of missed diagnosis and misdiagnosis.Method 1.A total of 95 patients with brucellosis diagnosed in the hospital of Inner Mongolia Medical University from February 2012 to December 2018 were retrospectively analyzed for the changes of peripheral blood and liver and spleen lymph nodes during hospitalization in patients with brucellosis.2.A total of 90 patients with brucellosis diagnosed in the Affiliated Hospital of Inner Mongolia Medical University and the Inner Mongolia Disease Control Center from September 2018 to December 2018 were selected as the study group, including 70 patients with acute phase(acute group).20 patients with chronic disease(chronic group). At the same time,30 healthy people in the physical examination were selected as the control group, and the fasting peripheral blood was taken in the morning.Flow cytometry was used to detect CD4+T,CD8+ T cells and CD4/CD8 in peripheral blood of patients with brucellosis and normal people. The values of the indicators were measured by immunoturbidimetric assay for the values of IgG,IgM,IgA,C3,C4,and C1 q in patients with brucellosis and normal subjects, and the changes in the above indicators were compared. Results 1.Of the 95 patients with brucellosis,22(23.1%)had leukopenia,4(4.2%)had elevated white blood cells,47(49%)had hemoglobin reduction, and 25(25.8%)had thrombocytopenia. There were 13 cases(13.7%)with a decrease in the three systems.Among 95 patients with brucellosis,87 patients underwent liver and spleen imaging examination, including 9 cases of hepatomegaly(10.3%),28 cases of splenomegaly(32.1%),and enlarged liver and spleen.6 cases(6.9%).Of the 95 patients with brucellosis,64 patients underwent lymph node imaging, including 7 withlymphadenopathy(9.9%).2.Compared with the normal group, the absolute counts of CD4+ T cells and CD8+ T cells in the peripheral blood of the disease group were higher than those in the normal group, and there was a statistically significant difference(P <0.01).There were no significant differences in IgG,IgM,IgA,C3,C4,C1 q,CD4/CD8 between the two groups(P > 0.05).Compared with the chronic group, the IgG,IgM,IgA,C3,C4,C1 q,CD4+T,CD8+T,and CD4/CD8 indicators were not significantly different between the two groups(P > 0.05).Conclusion 1.Brucellosis patients are prone to peripheral blood abnormalities and hepatosplenomegaly, need to be differentiated from blood system diseases and other bacterial or viral infectious diseases, reduce the rate of misdiagnosis and missed diagnosis, early diagnosis and treatment, prevention of chronic lesions.2.CD4+ T cells and CD8+ T cells increased during Brucella infection, and the cellular immune index changed significantly. However, there was no significant difference between the acute phase and the chronic phase.Cellular immunity may be the main mode of immunization for the prevention and elimination of Brucella.
