AIM:By determining the total sugar,total acid,amino acid nitrogen,PH value,alcohol content and other indicators of 11 batches of rice wine,the rice wine standard was established,and the draft of yellow wine was written to establish the fingerprint of yellow wine,and the stability and repeatability of rice wine were discussed.The processing techniques of earthworm and Earthworm Processed By Wine are preferred,and the optimal process is obtained by weighting the scores with hypoxanthine and inosine content as indicators.To explore the difference of pharmacological effects between different products of earthworm and Earthworm Processed By Wine,to study the antiasthmatic and antithrombotic effects of different products of earthworm,to improve the pharmacodynamic effects of earthworm,and to establish an effective method for processing the earthworm.METHODS:(1)The total sugar,amino acid nitrogen and total acid of rice wine were determined by titration;the alcohol content of yellow wine was determined by alcohol meter;the PH value of yellow wine was determined by PH meter;the calcium oxide content of rice wine was determined by inductively coupled plasma emission spectroscopy(ICP-AES).The content of methanol in rice wine was determined by gas phase;Establishment of Amino Acid Fingerprint of Yellow Wine by High Performance Liquid Chromatography.(2)The content of hypoxanthine and inosine in earthworm were determined by high performance liquid chromatography,and the best technique was analyzed by weighted score.(3)Taking the incubation period of asthma as the evaluation index,the experiment was carried out by using the drug spray-induced guinea pig asthma method to compare the high-dose and low-dose antiasthmatic effects of earthworm and its processed products.The mice were divided into 4 groups,which were blank group,positive drug group,earthworm group and Earthworm Processed By Wine group,through clotting time,tail bleeding time and induction of collagen-adrenalin.The effect of hemiplegia and mortality in the thrombosis model mice was to study the difference in antithrombotic effects of different processed products.SD rats were divided into 6 groups,which were blank group,model group,low dose group of raw materials,and raw materials.The high-dose group of earthworm,the low-dose group of Earthworm Processed By Wine,the high-dose group of Earthworm Processed By Wine,studied the antithrombotic effect of earthworm gun products by blood rheology experiment and the content of TXB2 and 6-keto-PGF1αin serum..RESULTS:(1)The total sugar content standard is set to 21.81-32.71g/L,the total acid content standard is set to 4.74-7.10g/L,the amino acid nitrogen content standard is set to 0.66-0.98g/L,and the PH value standard is set to 3.13-4.69,the alcohol standard is set to 12.8-19.2%,the non-sugar solids standard is 30.48-45.72g/L,the calcium factor standard is 249.17-373.76g/L,the methanol content should not exceed 0.04g/L,the fingerprint of rice wine The map has 19common peaks,of which five amino acids have been mapped,and the similarities are greater than 0.98.(2)The best processing technology of earthworm is to cut short section(5mm),wash for 2 minutes,and dry at 40℃.The best processing technology of Earthworm Processed By Wine is to add 12.5g of wine per 100g of medicinal material,simmer for 90 minutes and fry at140℃.The best pilot process of earthworm is take appropriate amount of original medicinal materials,remove impurities,cut into 58mm short sections on the cutting machine,rinse on the washing machine for 2min,and then take it out and dry in the oven at 60℃.The best conditions for the trial of Earthworm Processed By Wine is take 5kg of net dragon,add rice wine(every100kg of earthworm,add 12.5kg of rice wine),mix well,smother in a closed container for 90min,set the drum type speculum machine,220℃Heat,set the speed to 45 r/min,fry for 8 minutes,remove,spread,and let cool.(3)The guinea pigs that were sprayed with the same amount of 2%acetylcholine chloride and 0.1%histamine phosphate could cause the asthma latency to prolong(P<0.05 or P<0.01).And there is a very significant effect of relieving asthma.Among them,the low-dose anti-asthmatic effect of the Earthworm Processed By Wine is the best.From the clotting time,the tail bleeding time and the results of the thrombus model experiment,both the earthworm group and the Earthworm Processed By Wine group have antithrombotic effects.From the blood rheology experiment of the acute blood stasis model rats,compared with the model group,the whole blood reduction viscosity index of the local earthworm group was significantly reduced,and the TXB2 content in the serum decreased all the dragon groups.Both6-keto-PGF1αlevels were elevated.CONCLUSION:The quality standard of yellow wine was formulated,and the pass rate of each batch of 11 rice wines was more than 80%,which proved that the standard range of the subject was reasonable.After the system was made,the anti-thrombosis effect of earthworm was stronger than that of Earthworm Processed By Wine.a little.This project establishes a reasonable standard of yellow wine and establishes an effective processing technology,which is of great significance for the quality control of rice wine and earthworm.
