Cichorium intybus L.has the functions of clearing away liver and gallbladder,reducing stomach pain,eliminating heat and detoxifying.The main components of Cichorium intybus L.include polysaccharides,flavonoids,polyphenols and glycosides.Existing studies have shown that glycosides have many beneficial activities such as anti-bacterial,anti-oxidation,protection of cardiovascular and cerebrovascular,sedative and so on.But,little has been studied about the total glucosides of Cichorium intybus L.Therefore,fully exploiting and utilizing the resources of the plant of the Cichorium intybus L.,optimizing the process of extracting,purifying and synthesizing the total glycosides of the Cichorium intybus L.have provided the feasibility basis for further study and mass production of the total glycosides in the future,which have practical application to the comprehensive utilization of the Cichorium intybus L.and the development of the new product value.In this paper,we studied the extraction,separation and purification of the total glucosides of Cichorium intybus L.,the physical and chemical properties and the structure identification of the glucosides.The research process,results and conclusions are presented as follows:First,Response surface method?RSM?was used to design and optimize the condition of the technological process,which mainly focused on microwave extraction of total glycosides from Cichorium intybus L.Based on the single-factor experiment,comprehensive influences of 4 factors contributing to the yield of total glycosides was inspected respectively:concentration of ethanol,solid-liquid ratio,extraction time and extraction temperature.Based on the results of the single factor test,results showed that the optimal values of the above parameters were determined as follows:concentration of ethanol was 66%,solid-liquid ratio was 1:32?g/mL?,extraction time was 17 min,and extraction temperature was 77 0C.Under the optimized conditions,the experimental value of crude total glycosides yield was 6.139%which was in great agreement with the predicted value.The research offers a practical basis for the extraction of total glycosides from Cichorium intybus L.,the development as well as the utilization of the total glycosides.Then,the static adsorption-desorption experiments and adsorption kinetics of five macroporous resins for the purification of total glycosides from Cichorium intybus L.extracts have been evaluated.Equilibrium adsorption isotherms were constructed at different temperatures and the model of Langmuir and Freundlich was used to descibe the adsorption behavior of the selected optimal resin.Through the dynamic adsorption-desorption experiments,the process of purifying total glycosides by macroporous adsorption resin was established.The optimum conditions of HPD 300 was presented as follows:the sample concentration was 3.0 mg/mL,flow rate was 2.0 mL/min,the quantity of resin sample was26 mg/g and 30 mL 50%ethanol was used as eluant at the rate of 2 mL/min.With the optimum technological process,the collection ratio of total glycosides reached 75.79%and the purity reached 74.17%.Finally,the crude glycoside compounds were separated and purified by Cichorium intybus L..Two kinds of glycoside monomer compounds:A,B obtained through recrystallization were studied through IR,MS,1HNMR and 13CNMR respectively.The compared reference materials were cichorioside B and cichorioside C.
