Insect tea is an unique forestry resource insect product in China.It is made from the excrement of special insects feeding special plants.Orthopygia glaucinalis is one of the main insect-producing tea insects in Guizhou,and it is mainly distributed in southeastern Guizhou province.Its host vegetable oil fragrance tree and Cyclocarya paliurus.This article measured and evaluated the main nutritional components and health and medicinal value of Orthopygia glaucinalis-Cyclocarya paliurus insect tea.At the same time,this paper researchs on macroscopic and molecular expression of the anti-tumor effect of OCIT.The research on this aspect provides safe and effective,with independent intellectual property rights of tumor drug treatment to provide scientific theoretical basis and material.The main results of this paper are as follows.1.Analysis and Evaluation of Nutritional Components of OCIT.Analyzed the general nutrient composition,tea characteristic substance,amino acid,fatty acid,and mineral composition of the OCIT,and also evaluated the safety of the heavy metal content.As a reference to MLIT?ALIT and green tea,the nutritional value of the tea was evaluated.The results showed that the tea contains high tea polyphenols.Total amino acid content is higher,the eight kinds of essential amino acids accounted for about 39.3% of the total amino acids,essential amino acids and non-essential amino acids,the ratio of 64.76%,essential amino acids evaluation shows that grey tea amino acid nutritional model close to the egg and is better than that of soybean;The content of minerals is rich,which is the highest in Ca and rich in K elements,and the ratio of K/Na is 208.04.Heavy metal content is not exceeded.It is rich in amino acid and mineral content,complete in variety and reasonable in amino acid.It is a quality protein and safe mineral health drink with high nutritional value.2.In vitro anti-cancer activity assay of OCITMTT colorimetric assay was used to detect the inhibitory activity of grey green insect tea on the proliferation of MCF-7 breast cancer cells and SGC-7901 gastric cancer cell lines in vitro.The results showed that the grey green tea had good anti-cancer activity to the two cells.The inhibition rate for MCF-7 breast cancer cells was?95.18 ± 2.48?%,and the inhibition rate for SGC-7901 gastric cancer cells was?90.92 ± 2.27?%?90.92 ± 2.27?,for example,when the concentration of insect tea was 500 ?g·m L-1.The effect of grey green tea on the induction of apoptosis of the same tumor cells at different time periods was different.At the same time,the inhibition rate of Chinese insect tea on tumor cells was positively correlated with the concentration and time of treatment.For example,the inhibition rate of 48 h in 100?g·m L-1 to 500 ?g·m L-1 was?31.82 ± 2.32?% and?42.98 ± 1.89?.%,?54.97 ± 2.13?% and?64.78 ± 1.86?%.The ability of insect tea to induce apoptosis of different tumor cells was also different,for example,for human MCF-7 breast cancer cells,the IC50 of the tea was?322.17 ± 1.42?g m L-1 after 36 h,and the IC50 of the tea was?178.72 ± 0.94?g m L-1 after 48 h,but the IC50 of the human SGC-7901 gastric cancer cells after 36 h was?364.06 ± 0.97?.48 Tea IC50?220.35 ± 1.21?g.m L-1.This shows that the sensitivity of different tumor cells to insect tea is different.The reason is probably that different tumor cells are derived from different tissues and organs,and there are gene expression differences and different metabolic characteristics between them.3.Detection of Apoptosis-Inducing Apoptosis Mechanism of OCITReal time fluorescent quantitative PCR is one of the necessary tools for monitoring the markers of small residual molecular markers in tumor cells.The expression level of related apoptosis genes in MCF-7 breast cancer cells and SGC-7901 gastric cancer cells is detected by fluorescence quantitative PCR.The results showed that the expression level of Fas,Fas L,Bax and caspase in the MCF-7,Fas L,Bax and caspase family of grey green tea cells and SGC-7901 gastric cancer cells was obviously up-regulated.At the same time,to the anti apoptotic gene Bcl-2,grey green tea could significantly reduce the level of the expression of m RNA.Fas and Fas L as the exogenous apoptosis pathway related genes,Bax and Bcl-2 as endogenous apoptosis pathway related genes,and the experimental results show that the grey green tea can trigger both endogenous and exogenous apoptotic pathways in MCF-7 breast cancer cells and SGC-7901 gastric cancer cells,thus inducing apoptosis.At the same time effects on gene expression of gray green insect tea in a dose-dependent manner.
