Studies On Synthesis And Content Determination Of Ellagic Acid

Ellagic acid is a kind of polyphenols widely found in plants,which is often used in skin care and antioxidant.It has also been reported to play a certain role in anti-cancer,anti-tumor and anti-human immunodeficiency virus(HIV).In the early part of this century,the brookings institution for cancer research in the United States called ellagic acid “the only natural variety of cancer-fighting products available in the last decade.”In the 1950 s,the research on ellagic acid has been started abroad,and in the1980 s,ellagic acid has been applied in clinical trials abroad.Internationally,ellagic acid has been listed in the INCI(international standard catalogue of cosmetic raw materials),and China has listed ellagic acid in the Chinese name catalogue of international standards of cosmetic raw materials issued by the state food and drug administration(2010 edition).At present,there are many researches on the pharmacology of ellagic acid,but few reports on the industrial synthesis of ellagic acid.Most of the ellagic acid sold in the domestic market is from natural plants by extraction and purification,and the industrial production by organic synthesis is of low purity,which leads to high price of high purity ellagic acid.In this thesis,the synthetic process of ellagic acid was studied,and the route of high quality ellagic acid was explored.Based on the synthesis methods reported in the literature,we used two oxidation methods under alkaline and acidic conditions,taking methyl gallate and propyl gallate as raw materials respectively,and using a variety of oxidants to carry out the synthesis study.Our results showed that under acidic conditions,the yield of ellagic acid was the highest with relative content of 92% and absolute content of 89%.The purity of ellagic acid was analyzed.The determination method of ellagic acid was established by HPLC method.The linear regression equation of ellagic acidwas y=10186x+21705(R=0.9998,n=7).The applicability,reliability and scientificity of the analytical conditions were investigated.In addition,the biological activity of mononuclear macrophage RAW264.7 was preliminarily studied for ellagic acid and its analogues,in order to understand its anti-inflammatory activity.
